| 中文摘要 | 停經婦女經過荷爾蒙補充療法後,心臟血管疾病發生率明顯降低。因此雌性素可能具有調控血壓、保護心臟血管功能的作用,本實驗選用雌性SD大白鼠並以去卵巢模擬類似人類停經的動物模式,探討缺乏E2及去卵巢再長期補充E2治療,對血管反應性及血流動力學的影響及其可能作用機轉。從實驗結果顯示,雌性大白鼠接受卵巢切除術(ovariectomy, Ovx)後,其體重及左心室的重量,明顯比接受手術後再施予長期雌二醇治療(Ovx+E2)組及假手術對照組(sham)為重,但子宮卻呈萎縮現象,且血中17β-estradiol的含量均顯著降低(P<0.05)。在血流動力學方面,結果顯示,Ovx組之心搏出量、心輸出量明顯下降,但心跳及週邊血管阻力顯著上升(P<0.05),經長期補充E2治療後,上述變化參數均恢復至正常。以上的實驗報告顯示雌性素缺乏的主要作用是在增加總週邊血管阻力。 |
| 英文摘要 | Menopause women after estrogen replacement therapy will significantly reduce the incidence of cardiovascular disease. Therefore estrogen has real effects to control blood pressure and protect cardiovascular function. These experiments were conducted by using ovariectomized SD rats as a model of menopause women with estrogen deficiency. We studied the effect and possible mechanism of estrogen on cardiovascular hemodynamic changes. The results showed that female SD rats after ovariectomy, body weight significantly increased and developed left ventricular hypertrophy, but uterus was smaller and plasma content of 17β-estradiol was significantly decreased (P<0.05) when compared to sham and Ovx+E2 group. The study of steady hemodynamic changes revealed significant decreases of stroke volume and cardiac output, and significant increases of heart rate and total peripheral resistance (P<0.05) in Ovx group. These results indicate that the major consequence of strogen deficiency is due to the increment of total peripheral resistance. |
