中文摘要 | 去甲斑蝥素(Norcantharidin, NCTD)是人工合成的一種新型抗癌藥物,為中藥 斑蝥(Mylabris)體內抗癌有效成份斑蝥素衍生物。在臨床應用上,已有去甲斑蝥素治療原 發性肝癌的報導。但去甲斑蝥素對癌細胞抑制作用的詳細機轉尚未知,同時斑蝥的另一衍生 物去水去甲斑蝥素(Dehydronorcantharidin, DNCTD)對肝癌細胞株的影響經由實驗,其亦 會造成肝癌細胞株的死亡,只在於所需藥物濃度的不同。DNCTD在HA22T肝癌細胞株中約需10 位的NCTD劑量,在Huh7肝癌細胞株中約需5倍的NCTD劑量。時間的因素方面,NCTD在24小時 即引發細胞凋亡的作用,DNCTD則需72小時才有作用。至於其造成的影響,經由本實驗,由 細胞之變化與Flow cytometry分析等,已可証實其作用為Apoptosis。至於其真正機轉尚需 作進一步的分析探討。 |
英文摘要 | Norcantharidin (NCTD) is a demethylated form of cantharidin with antitumor properties. It was used as a routine anticancer drug against hepatoma. However, there is limited study on the effect of NCTD on human cancer cells, caused progressed to apoptosis within 48 hours in two human hepatoma cell lines: HA22T and Huh-7. The precise mechanism of NCTD is still unknown. Dehydronorcantharidin (DNCTD), is another demethylated form of cantharidin with antitumor properties that caused progressed to apoptosis within 72 hours. DNCTD need 10 times dose than NCTD induced apoptosis in HA22T, and need about 5 times dose in Huh7 cell lines. However the mechanism of anticancer is unknown, still need further investigation. |
