英文摘要 | The purpose of this experiment was to isolate and purify respiratory macrophages of the duck, and the cell morphology under scanning electron microscope also investigated. Duck respiratory tract macrophages were elicited with a 1:4(v/v) suspension of imcomplete Freund's adjuvant in a sterile phosphate-buffered saline by injection directly into the abdominal air sacs. Respiratory tract macrophages were collected on days 0, 1, 2, 3, 5, 7 and 9 after injection. The macrophages were then purified by passage through a Ficoll-Hypaque gradient resulting in 96.3± 10.13% purity and 95.2±5.4% viability. On days 7 and 9 after injection, maximal numbers (6.85±4.32×10□ macrophages per duck) of respiratory tract macrophages for in vitro experiments were obtained. The macrophages demostrated the ability to attach to cover glass and to kill Escherichia coli in vitro. Image of scanning electron microscope showed the cell wtih a highly-folded cell membrane and flat or filamentous pseudopods. |
