中文摘要 | 本研究以GC-MS-SIM建立了一套檢驗尿液中睪固酮及表睪固酮的分析方法,以d3-睪固酮為內標準品,睪固酮濃度為20ng/ml~300ng/ml時,所得標準曲線迴歸方式為y=0.002x + 0.054 (r=09894); 表睪固酮濃度為20ng/ml~300ng/ml時,所得標準曲線迴歸方程式為y=0.001x + 0.027 (r=0.9967)。睪固酮萃取之回收率平均為68.0%。以β-glucuronidase酵素水解。以N-methyl-N-(trimethylsilyl)-trifluoro acetamide衍生化。睪固酮之滯留時間為14.35分,表睪固酮之滯留時間為13.91分,質譜儀則以指定離子監測方式。本法可分離與定量尿液中所含之睪固酮與表睪固酮。 |
英文摘要 | A GC-MS-SIM method was developed for the analysis of testosterone and epitestosterone in urine samples. The internal standard used was d3-testosterone. The linear calibration range was 20~300 ng/ml for both compounds (r=0.9894 for testosterone). The mean of recoveries was 68.0%. Beta-glucuronidase was used to digest the conjugated form of testosterone metabolites. N-methyl-N-(trimethylsilyl)-trifluoro acetamide was used to derivatized. The retention time was 14.35 min for testosterone, and 13.91 min for epitestosterone. The results indicated that the proposed method is a suitable method for the determination of testosterone and epitestosterone in urine. |
