中文摘要 | 肝素具抗凝血及抗脂血活性,是治療冠狀動脈疾病用藥。本研究利用生物降解性材料--幾丁聚醣包覆肝素,製備成肝素緩釋放小球;同時探討幾丁聚醣特性對包覆肝素之包覆效率、表面型態及釋放趨勢之影響。研究結果顯示在固定肝素濃度下,包覆效率會隨幾丁聚醣分子量與微小球粒徑增大而增加;另外,肝素總包覆量有飽和現象的產生,與微小球表面肝素結晶之產生相吻合。釋放測試實驗發現釋放速率相當快速,且於釋放終止時約有40%的肝素殘留於微小球內部。為了降低快速釋放的現象,以1.5%幾丁聚醣進行微小球雙層包覆實驗,證實雙層包覆之微小球可達成肝素緩釋放之系統。 |
英文摘要 | Heparin was recommended in treatment of patients with acute coronary syndrome. To delivery heparin with sustained releases, encapsulated heparin by chitosan microspheres, a biodegradable biomaterial, has been tried and investigated. In this study, the effects of the characteristics of chitosan on encapsulation efficiency, morphology, and release profiles of the heparin loaded chitosan microspheres have been studied. The results showed that encapsulation efficiency of heparin was increased with molecular weight of chitosan to form microspheres and their sizes at a fixed heparin concentration. Moreover, saturated phenomena of heparin loaded in microspheres were found, and consistent with the observation of heparin crystals on the surface of the microspheres. The initial burst and fast release of heparin from microspheres was observed in all cases while 40% of residual heparin within the microspheres was also found at the highest concentration of loaded heparin (e.g., 500 iu/ml). To reduce those fast release profiles of heparin, double-layer coating the microspheres with 1.5% of chitosan worked well. Through the study the characteristics of chitosan affected the characteristics of heparin loaded chitosan microspheres and double-layer coating the microspheres could sustain the releases of heparin. |
