中文摘要 | 截至民國89年底,臺北市立動物園共有25隻黑猩猩,其中有五隻為小於兩歲半的新生個體。為了確定這些新生個體及其他黑猩猩的親緣關係,以方便管理及避免近親交配所衍生出來的問題,本研究利用分子生物學的方法來鑑別個體的交替基因型以求確認其親子關係。嘗試以人類法醫檢驗最主要之十三型STR(Short Tandem Repeats, 短片段重覆性基因)系統及一項性別系統,運用於黑猩猩的檢體,結果發現於黑猩猩可獲得STR十型及性別基因型的結果。亦即萃取白血球細胞的DNA,再利用十四組不同的核酸引子(primer)將體染色體上的十三個基因座及性染色體上的一個基因座區域之DNA片段增殖出來,再以自動化分析儀及軟體分析各片段的長度並進一步判定其型別。其中有三個STR基因座懷疑因引子專一性不足,所以無法應用在黑猩猩。其他的十個STR基因座及一個性別基因座上的DNA片段皆可在黑猩猩上被增殖出來。藉由此25隻黑猩猩個體STR多型性資料庫的建立及基因型的比對,除了一隻出現無法判定之特殊型別外,此族群彼此之間的血緣關係皆已確認;並藉此進一步推測另一隻已歿的黑猩猩酋長的基因型。由此可知人類STR十三型及性別基因型系統可應用在黑猩猩血緣關係的鑑定,其將有助於管理、育種及繁殖計畫的進行。 |
英文摘要 | Until late 2000, 25 chimpanzees were raised at Taipei City Zoo. Among these, there were five under two and a half years old. To make sure of the blood relationships of these newborns and others in order to manage easily and avoid inbreeding, we use molecular biology method to identify individual genotypes. We tried to make use of human medicoleagal STR and sex system on the chimpanzee and finally got 10 STR loci and sex allelic genotypes. We extracted DNA from the white blood cells and used 14 pairs of different DNA primers to amplify 13 STR loci on autosomatic chromosome and one locus on sex chromosome. These amplified DNA segments were analyzed by automatic analysis equipment and software to determine the lengths and the genotypes. Among these 14 loci, 3 STR could not be typed on the chimpanzees, which maybe due to not enough specificity of the primers. However, each DNA fragment of the other 10 STR loci and sex locus could be amplified on the chimpanzee samples. According to the obtained STR polymorphism data of these 25 chimpanzees and allelic genotypes comparison, we determined all of the relationships in this population, except one chimpanzee. Moreover, based on the STR polymorphism data of this population, we could infer the genotypes of another dead chimpanzee, Chiu-Chang. From this research, we found that human 13 STR and sex genotyping system could be applied on the relationship identification of chimpanzees, and it will be helpful to the management and the proceeding of breeding program. |
