中文摘要 | 本研究比較鐮孢菌13種分化型的隨機增幅核酸多型性(Random amplified polymorphic DNA; RAPD),在以KS、P2與P6引子增幅所得的多型性圖譜中,發現由P6引子擴增尖鐮孢菌蘿蔔分化型(F. o. f. sp. raphani)時,得到一個獨特的DNA片段,將此1,045 bp的片段製備成探針P61045r,與RAPD產物進行雜合反應時,僅尖鐮孢菌蘿蔔分化型具有正反應。由RAPD衍生具分化型專一性的探針,可應用於區分尖鐮孢菌蘿蔔分化型。該探針的核酸序列與尖鐮孢菌埃及豆分化型由引子P6增幅出的一段DNA有98.5%的相似度,埃及豆分化型的該段基因被認為是黃化相關蛋白的基因。尖鐮孢菌蘿蔔分化型的這段基因是開放讀取架構,但其功能尚待了解。由於這二段DNA的高相似度,探針P61045r 應無法區分這二個分化型,但台灣尚未有尖鐮孢菌埃及豆分化型報導。 |
英文摘要 | Random amplified polymorphic DNA was used to characterize different pathotypes of Fusarium oxysporum. When a 1,045-bp DNA fragment amplified from F. o. f. sp. raphani isolate with primer P6 was used as probe P61045r , it hybridized with RAPD product derived from F. o. f. sp. raphani only. The RAPD-derived, forma specialis-specific probe may be used to distinguish isolates of F. o. f. sp. raphani. The nucleotide sequence of the RAPD marker P61045r for F. oxysporum f. sp. raphani had 98.5% identity with a DNA fragment generated by primer P6 of F. o. f. sp. ciceris, accession number AF005634. The RAPD fragment was described as a F. oxysporum yellowing-associated protein gene. The same gene we amplified from F. o. f. sp. raphani with primer P6 possibly was an open reading frame. While the function of this gene need more evidence. Fusarium o. f. sp. ciceris has not been reported in Taiwan and the isolates were not included in the study. However, with 98.5% homology to F. o. f. sp. ciceris, it would be very difficult to discriminate F. o. f. sp. raphani from F. o. f. sp. ciceris by the probe. |
