中文摘要 | 本研究利用降解速率、生物相容性分析和機械性質來探討戊二醛、雙環氧化物、水溶性氰胺物等三種交聯劑對動物明膠支架的影響,發現水溶性氰胺物為交聯動物明膠最佳交聯劑。接著將幾丁聚醣和動物明膠以不同比例混合,並以水溶性氰胺物交聯,進行各種測試。在降解速率測試中,幾丁聚醣幾乎不降解,而在1:1比例混合物中的動物明膠降解最慢,再經由接觸角、生物相容性、機械性質和孔洞結構等測試後,發現其三維支架結構最穩定且生物相容性亦最佳。因此幾丁聚醣和動物明膠在1:1比例混合物在所有幾丁聚醣和動物明膠混合物中,適合用來製作軟骨再生支架。 |
英文摘要 | In this study, the chitosan-gelatin complexes were evaluated as tissue engineering scaffolds for cartilage regeneration. At first, a better crosslinker for gelatin was selected among glutaradehyde (GA), bisepoxy (E) and water-solube carbodiimide (WSC), base on the analysis of degradation rate, biocompatibility and mechanical properties of the crosslinked scaffolds. WSC was found to be the most crosslinker. Later, a series of the chitosan-gelatin complexes crosslinked by WSC were created. With cmtosan-gelatin ratio equal to 1, the complex had the lowest degradation rate, demonstrating an interaction between these two components. Evaluation based on contact angle, chondrocyte proliferation, as well as the pore structure and stability of the scaffolds led to a conclusion that such complex could be used to fabricate the tissue engineering scaffolds for cartilage regeneration. |
