中文摘要 | 利用大鼠肝臟細胞之初代培養偵測食品外源物質之細胞毒性,培養4小時呈穩定狀態時之細胞分別以食品真菌毒素及天然食品添加物或天然香科培養,培養濃度分別為0.01~2.50μg/ml及0.02~7.50μg/ml,經培養24小時測定系列肝臟細胞受毒害後之酵素釋出及功能指標,藉以評估外源物質對初代培養大鼠肝臟細胞之毒害活性,經培養後出現乳酸脫氫?(LDH),天冬氨酸轉胺?(AST)及丙氨酸轉胺?(ALT)之釋出顯著增加(p<0.05),細胞內蛋白質及細胞存活率之顯著減少(p<0.01)及細胞病理效應之異常增加等。這些測試之外源物質之細胞毒性效應均出現顯著之劑量-依賴性關係,其毒性依序分別為黃麴毒素B1>黃米毒素>伏馬鐮胞毒素B1>嘔吐毒素;槲皮酮>堪非醇>甘草素>咖啡因;黃樟素>金絲桃蔥酮>香豆素>小蘗鹼>山道年>細辛醚>蘆薈素及蒜素。在本試驗中測試物質對肝臟細胞毒性之閾下劑量分別為黃麴毒素B1-0.01μg/ml;伏馬鐮胞毒素B1,黃米毒素及嘔吐毒素-0.05μg/ml;黃樟素-0.10μg/ml,堪非醇,槲皮酮,金絲桃蔥酮,香豆素及小蘗鹼-0.50μg/ml;甘草素,細辛醚及山道年-1.50μg/ml;咖啡因-7.50μg/ml;蘆薈素及蒜素除於劑量為7.50μg/ml時乳酸脫氫?之釋出有輕微之增加現象外,其餘雖然與其它天然香料授予相同之劑量但均未出現顯著之細胞毒性指標。另研究顯示以大鼠肝臟細胞代謝完整性之偵測,在天然外源物質毒性評估之特殊偵測範圍,如細胞受到傷害之後期乳酸脫氫?(LDH),天冬氨酸轉胺?(AST)及丙氨酸轉胺?(ALT)之釋出,細胞內蛋白質含量,細胞存活率及細胞之病理效應等是敏感性大之偵測重點項目。 |
英文摘要 | Primary cultures of rat hepatocytes served as an usual alternative model for evaluation of the cytotoxicity of xenobiotics. Cellular injury was assessed by means of a series of enzymatic and functional indices in 4-h-old cultures exposed for 24 h to various food mycotoxins ranging from 0.01 to 2.50 μg/ml, natural food additives and natural aromas ranging from 0.02 to 7.50 μg/ml, respectively. In cultures, clear evidence of cytotoxicity was observed as evaluated by leakage of LDH, AST and ALT, which significantly increased (p<0.05) while the intracellular protein and cell viability significantly decreased (p<0.01). Meanwhile, the cytopathic effects clearly displayed a dose-dependent increase. The cytotoxic effects of these tested natural xenobiotics clearly occurred in a dose-dependent manner and in the order of aflatoxin B1>luteoskyrin>fumonisin B1>deoxynivalenol; quercetin>kaempferol>glycyrrhizin>caffeine; safrole>hypericin>coumarin>berberine>santonin>β-asarone>allicin or aloin. In this study, the thresholds of cytotoxic concentration were 0.01 μg/ml aflatoxin B1,0.05 μg/ml fumonisin B1, luteoskyrin and deoxynivalenol; 0.10 μg/ml safrole, 0.50 μg/ml of kaempferol, quercetin, coumarin, hypericin and berberine, 1.50 μg/ml glycyrrhizin, β-asarone and santonin and 7.50 μg/mlcaffeine. The cytotoxicity was not remarkably effected by allicin and aloin in the same concentration as the other tested natural aromas. Furthermore, LDH leakage slightly increased at a dose of 7.50 μg/ml. These findings also indicate that measures of metabolic integrity of cultured rat hepatocytes are sensitive indices of specific measures for evaluation of the cytotoxicity of natural xenobiotics, such as the leakage of LDH, AST and ALT, intracellular protein content, cell viability and cytopathic effects of cultured rat hepatocytes, which can detect the later stages of cell injury. |
