英文摘要 | Under 70μmol-m-2.sec-1 photosyntetic photon flux density (PPFD), explants of Rosa hybrida Hort. cvs. Samantha and Landora were cultured in containers enriched with carbon dioxide of 2000 ppm for 15 min. After 75min, the concentration of carbon dioxide in the contaier was 223 or 211 ppm, respectively, when ‘Samantha’ explants were cultured on the medium containing 15g/1 or 7.5g/1 surcrose. ‘Landora’ explants were cultured on the same medium above, the concentration of carbon dioxide in the container was 316 or 140ppm respectively. Under 70μmol-m-2.sec-1 PPFD, for 2 hours, culture containers were pumped carbon dioxide of 2000 ppm for 15min at the beginning of every 2 hours. The explants of ‘Samantha’ and ‘Landora’ had the heaviest dry weight and the best performances when they were cultured on the medium containing 7.5g/1 sucrose. Microcuttings were inserted under mist for 3 weeks. Microcuttings of ‘Samantha’ and ‘Landora’ from containers enriched carbon dioxide rooted well than those from containers without carbon dioxide enrichment. In addition, with carbon dioxide enrichment, ‘Landora’ explants grown on the medium containing 7.5 g/l sucrose were rooted well than those grown on the medium containing 15g/l sucrose; the former microcuttings had more lateral roots. The method of carbon dioxide enrichment for acclimatization in vitro was also discussed. |
