中文摘要 | 前面的研究顯示並沒食子酸(Ellagic acid)抑制了老鼠血液和肝臟組織 N-乙醯轉移酵素活性。並沒食子酸(Ellagic acid)被用來檢測對老鼠膀胱和大腸 組織N-乙醯轉移酵素的影響。以高壓層析儀來檢測而決定N-乙醯轉移酵素對芳 香胺類2-Aminofluorene的乙醯化和非乙醯化的量來量出該酵素的活性。隨著並 沒食子酸(Ellagic acid)劑量增加,老鼠膀胱和大腸組織N-乙醯轉移酵素活性也 跟著相對的減少,也就是說並沒食子酸濃度增高則膀胱和大腸組織中該酵素活性 則愈低。結果同時也顯示並沒食子酸降低老鼠膀胱和大腸NAT酵素的Km和 Vmax值。這是第一次報告並沒食子酸抑制老鼠膀胱和大腸NAT酵素活性. |
英文摘要 | Our previously studies had shown ellagic acid inhibited N-acetyltransferase (NAT) activity in Sprague-Dawley rat blood and liver tissues cytosols. In this study, ellagic acid was selected for determining the effect of arylamine NAT activity in rat bladder and colon tissue cytosols. By using high performance liquid chromatography, NAT activity for acetylation of 2-aminofluorene and remain unacetylated 2- aminofluorene were examined. The NAT activity in rat bladder and colon tissues was inhibited by ellagic acid in a dose-dependent manner: i.e. the greater the concentration of ellagic acid in the reaction, the greater the inhibition of NAT activities in both examined tissues. The data also indicated that ellagic acid decreased the apparent Km and Vmax of NAT enzymes from rat bladder and colon tissues cytosol. This findings is the first demonstration which showed ellagic acid effect rat bladder and colon tissues NAT activity. |
