中文摘要 | 建立了Bt殺蟲晶體蛋白的雙抗體夾心ELISA檢測方法,檢測限達1ng/mL以下。以此方法檢測了轉Bt基因抗蟲楊樹株系12、153、192葉片中ICP含量,結果與其他檢測方法所得一致,與對楊尺蠖和舞毒蛾的校正死亡率呈顯著正相關,是生產上定量檢測抗蟲性的較好方法。不同年齡葉片中,以ngBt蛋白/gFW葉片表示的ICP含量有差異:展開葉>幼葉>成葉>老葉,而1CP含量占可溶性蛋白比例相對穩定。與對照株系相比,轉基因楊樹葉片中可溶性蛋白含量較低,生長素(IAA)、玉米素及其核?(Z+ZR)、赤黴素(GAs)含量降低,而脫落酸(ASA)含量提高,可能是轉基因楊樹葉片較小、生長勢弱的生理原因。 |
英文摘要 | A double sandwich EL/SA with monoclonal and polyclonal antibodies to CryIA (c) was established, of which the detection sensitivity was less than I ng/mL. Bacillus thuringiensis (Bt) insecticidal crystal protein (ICP) in leaves of three transgenic poplar (Bt-poplar) varities, Populus nigra 12, 153, 192 were measured using, the results were similar with other methods. The contens of ICP content in leaves of Bt-poplar were posive corelated to corrected mortality for Apocheimia cinerarius Erscholf and Lymantria dispar Linnacus. The contens of ICP content in fresh wight of leaves were different with leaf age, while in soluble protein were stable. Compared with control poplar, the contents of soluble protein in Bt poplar were lower, all the contents of auxin(IAA), cytokinins (Z+ZR) and geberi!ins (GAs) were decreased, hewer abscisice acid (ABA) content was increased, which might be related to small leaf and slow growth of Bt poplar plants. |
