英文摘要 | A novel enzyme purification technology, expanded bed adsorption (EBA), has been developed for efficient direct recovery of α -glucosidase from unclarified baker's yeast cell homogenate. STREAMLINE ion exchange and hydrophobic interaction adsorbents were chosen for the EBA purification process. For improving purification efficiency, optimal operating conditions were determined in a series of small scale batch and packed bed experiments. These experiments included the choice of adsorption pH, the effect of ionic strength for the adsorption process, the study of dynamic binding capacity, and the choice of elution conditions. Ion exchange and hydrophobic interaction adsorbents were compared for their purification performance. The results showed that both adsorbents for the direct recovery of activity of α -glucosidase by using expanded bed chromatography were approximately 80%. However, the purification factor was only two-fold. On the basis of these results, the relative productivity of ion exchange operating mode was higher than that of hydrophobic interaction operating mode. Hence, STREAMLINE ion exchange adsorbent is more suitable for use in large scale purification processes. |
