| 中文摘要 | 本研究針對非極性之ODS分離管柱(Octadecylsilyl column)的分離效率做一探討,以三種不同廠牌(代號A、B及C等)之ODS分離管柱的分離進行評估。實驗分為二部分,一為探討ODS填充之良否,以理論板數及不對稱性來進行評估,使用之動相為蒸餾水和甲醇之混合溶液(40/60, v/v),分析樣品為甲苯。另一部分為以兩種不同pH之磷酸緩衝液(pH2.5及pH7.0)為動相,並分別以benzyl alcohol、acetylsalicylic acid和N, N,-dimethylaniline等四種不同解離型態之化合物為分析樣品,進行矽膠上-OH是否鍵結完全之測試。結果顯示管柱之理論板數大小為A>C>B;層析峰不對稱性之優劣順序亦依序為A>C>B,顯示管柱之填充緊密度與管柱分離效果(對稱性)呈一致性。矽膠上C18之鍵結程度,則以B、C為佳,A管柱稍低。 |
| 英文摘要 | The objective of this study is to evaluate the resolution efficiency of HPLC column, specifically for ODS column (Octadecylsilyl column or RP-18 column). Silica gel was generally used as the packing material in column, however, alkyl group (C18) were chemically bounded to the OH group of silica gel to extend its multi-functions. Three different brands of ODS column (Code No. A、B and C) were used as the study materials. To evaluate the perfectness of packing technology, theoretical plate numbers and peak asymmetry tests were used as the criteria. Mixture of distilled water and methanol (40/60, v/v) was used to be the mobile phase, and toluene was the analytical sample. To investigate the completeness of C18 coverage, mixture of phosphate buffer (pH 2.5 and pH 7.0) were used as the mobile phase. Benzyl alcohol、caffeine、acetylsalicylic acid and N. N.-dimethylaniline were used as the analytical samples. Theoretical plate numbers indicated that A>C>B, whereas the same tendency were also shown in the peak asymmetry test. The results of C18 coverage showed B and C were both tallies with the recommended standards, but of that, was not shown in A column. |
