中文摘要 | 三週齡仔豬 16 頭分別飼以四種不同日糧脂肪-椰子油、魚油、豬油以及大豆油,以改變呼吸道發炎,介質-白三烯素(leukotriene;LT)、類前列素(prostacyclin;PG)及類前凝素(thromboxane;TX)的生成,同時測定胸腺與脾臟的白血球中介質Ⅳ(interleukin-4;IL-4)、白血球中介質Ⅷ(interleukin-8;IL-8)及干擾素-伽瑪(interferon-gamma;IFN-γ)的基因表現,以探討仔豬免疫組織細胞素基因表現對發炎介質生成的調節作用。結果顯示不同日糧脂肪改變肺組織脂肪酸組成而影響呼吸道發炎介質的生成,魚油顯著地減少PG及TX的生成(P<0.05),但對於LT沒有相同的效益。日糧脂肪對胸腺與脾之IL-4及IFN-γ基因表現並無顯著影響。然而,LT的生成量與脾IL-4之基因表現呈顯著地正相關(r=0.80,P=0.0006);刺激原存在下,LT的生成量則與脾IL-8之基因表現呈正相關之趨勢(r=0.60,P=0.057);而脾IL-4與IFN-γ的基因表現則為負相關(r=-0.55,P=0.025);脾IL-8之基因表現與脾IFN- γ之基因表現亦呈負相關之趨勢(r=-0.50,P=0.055)。此實驗結果顯示仔豬呼吸道白三烯素的生成與脾臟IL-4、IL-8基因表現有一致的趨勢,而脾臟IL-4及IFN-γ則為反應相反之基因表現。 |
英文摘要 | To investigate the interactions between cytokine gene expressions and eicosanoid synthesis, 16 piglets were randomized into four groups and fed diets containing 3.6% coconut oil, fish oil, lard, or soybean oil plus 0.4% soybean oil for 21 days to modulate the synthesis of eicosanoids including leukotriene (LT), prostacyclin (PG), and thromboxane (TX) in the lungs. Interleukin-4 (IL-4), interleukin-8 (IL-8), and interferony-γ (IFN-γ) gene expressions in spleen and thymus were examined using RT-PCR. The results showed that fish oil effectively inhibited the synthesis of PG and TX (P<0.05) in lungs but did not reduce the synthesis of pulmonary LT. Interleukin-4, IL-8, and IFN-γ gene expressions in the spleen and thymus were not affected by dietary fat. However, a significant positive correlation was observed between LT synthesis in the lung and IL-4 gene expression in the spleen (r=0.80, P=0.0006). Stimulated-LT production in lungs tended to be positively correlated to splenic IL-8 gene expression (r=0.60, P=0.057). Negative correlation was found between IL-4 and IFN-γ gene expressions in the spleen. The mRNA levels of IFN-γ were also negatively correlated to those of IL-8 in the spleen. Therefore, the positive correlation between splenic IL-4 gene expression and the release of LT in the lungs was consistent with clinical features in asthmatic patients. In contrast, the negative correlation between IL-4 and IFN-γ gene expressions in spleen idicated that IFN-γ and IL-4 gene expressions in the spleen were apparently converse. |
