中文摘要 | 將高約 5cm 臺灣金線連( Anectochilus formosanus Hayata ),栽培於無菌聚 丙烯( polypropylene )塑膠袋,調查幼苗期以後金線連的生長, 結果顯示只添加培養液 而不加其他介質的生長最佳。當培養液蔗糖濃度由 2% 增加至 3%,莖部與全株的乾重 / 鮮 重之比分別增加 37.6% 與 25.8%,類似效果亦出現在含 0.3% 花寶 1 號( HYPONeX No.1, N-P-K, 7-6-19 )及 0.15% 酵母抽出物的培養液。 2% 蔗糖比 3% 蔗糖培養液顯著地提高葉 / 莖部之乾重比, 亦提高葉部乾物率 / 莖部乾物 率之比,此顯示在無菌狀態下,碳源供應強度會改變植株內碳水化合物的分配。 3% 蔗糖比 2% 蔗糖培養液可提高植株葉片總醣含量、並降低葉片的澱粉含量及還原醣含量。 比較上述 之密閉系統培養與生長箱之開放系統培養, 密閉系統有利於提高乾重 / 鮮重比、莖部生長 、促進葉部還原醣,總醣及澱粉的累積。 |
英文摘要 | The post-seedling plants of A. formosanus Hayata, one of the rare medicinal herbs in Chinese traditional medicine, were grown under sterile condition. Sterile-cultured plants about 5 cm hight were transplanted into sterilized polypropylene-made plastic bag. The results indicated that the plants received in liquid medium only showed the best growth responses. Dry weight/ fresh weight ratio on stem and shoot of the plant was increased by 37.6% and 25.8% respectively, when the sugar concentrations were increased from 2% to 3%. Similar effect were also found in media containing 0.3% HYPONeX No.2 (N-P-K, 7-6-19) and 0.15% yeast extract. The ratios on dw□/dw□ and leaf□/stem□ of plants cultured in 2% sucrose-media were significantly grealer than that of 3% sucrose-media. It appeared that the intensity of carbon supply will change the partition of dry matter in plant under sterile condition. Total sugar content of leaf in plants cultured in 3% sucrose-media was higher than that of 2% sucrose-media. On the other hand, the starch content and reducing sugar content of the plants cultured in 3% ssucrose, was lower than that of the plants cultured in 2% sucrose. Comparing closed-culture system as mentioned and open-culture system in growth chamber, the former had the advantages in increasing ration on dw/fw, stem growth, and promoting accumulation of reducing sugar, total sugar and starch content in leaf. |
