中文摘要 | 由近來的研究中,研究人員發現基丁寡糖 (chitooligosaccharide) 對動物有激發免疫系統和抑制癌細胞生長的生理活性。一般而言,製造基丁寡醣的方式需要濃酸及高溫來部份水解基丁質。此製程不但產生大量廢水且危險。因此必需開發更可行的方法。本研究的目地是發現及純化基丁質分解酵素來部份水解基丁質以得到基丁寡醣。這個研究報告中,我們由一株培養於只有以基丁質為唯一碳源的細菌,利用離子交換樹脂、分子篩膠過濾、疏水性作用管柱層析,純化了二種基丁質分解酵素。這二個基丁質分解酵素分別為外切基丁質分解酵素 (exo-chitinase 和雙切基丁質分解酵素 (chito-biosidase),其分子量經SDS-PAGE電泳測定分別為28及60KDa。 |
英文摘要 | Recent studies of the treatment of animals with chitooligosaccharides have demonstrated that they can activate the immune system and suppress tumor growth. Heretofore, in order to obtain partially hydrolyzed chitin for producing chitooligosaccharides, a reaction had to be initiated utilizing concentrated acids and high temperature. A less complicated process is preferred. Therefore, we aimed to identify and purify chitinases that are capable of producing chitooligosaccharides enzymatically. Thus two chitinases were purified from bacterial cultures when the carbon source was limited to colloidal chitin prepared from crab shell. The chitinases were purified to near homogeneity by ion-exchange, gel filtration, and hydrophobic interaction column chromatography. The molecular weight of the purified chitinases was determined to be 28kDa and 60kDa, based on SDS-polyacrylamide gel electrophoresis analysis. The two chitinases were determined to be exo-chitinase and chitobiosidase. |
