中文摘要 | 鴨、鵝等水禽類小病毒所引起之致死性腸炎,其臨床症狀及剖檢常與多種疾病混 淆,鑑別診斷往往須要等待較費時之傳統病毒學檢驗結果。 本實驗即利用聚合醣連鎖反應 (PCR) 之原理,得以直接由感染臟器樣品中偵測出水禽類小病毒 DNA,達成快速且正確診斷 之目的。 我們將收集自本省之鴨、鵝小病毒分離株作初步的鑑識, 以特異性引子進行 PCR 反應來增幅其結構蛋白基因, PCR 產物經核酸序列定序分析, 並與基因庫之禽類小病毒 DNA 序列進行比對,確認為水禽類小病毒株。結果顯示由病材肝臟乳劑樣品中之陽性檢出率 (65.38%) 最高。 |
英文摘要 | Waterfowl parvoviruses cause dreadful disease in goslings and ducklings The clinical signs and necropsy findings are similar to many other diseases. Therefore, differential diagnosis usually relies on traditional laborious virological tests. This study aims to establish a rapid and specific method for direct detection of viral DNA in infected tissues by polymerase chain reaction (PCR). PCR reactions were performed with specific primer to amplify the structural protein gene from specimens collected from infected ducks and geese in Taiwan. Sequences of PCR products were confirmed to be parvovirus after comparison with the data in the gene bank. Our data revealed that emulsion from infected liver provide a high detecting rate (65.38%) for this disease. |
