中文摘要 | 本研究利用acid phosphatase、alkaline phosphatase、catalase、diaphorase、esterase、formate dehydrogenase、s-glucosidase、glutamate dehydrogenase、glutamate oxaloacetate transaminase、malate dehydrogenase、malate enzyme、peroxidase及shikimate dehydrogenase等13種同功異構酵素,以聚丙烯銨膠體電泳技術分析尾孢菌及假尾孢菌之電泳型(electromorphs),依據此酵素電泳型將Cercospora arachididicola (CaP)、C. canescens (CcC與CcM分別採集自豇豆與綠豆)、Pseudocercospora bradburyae (PbC)及P. cruenta (PcC1、PcC2、PcS及PcA分別採集自豇豆、菜豆及紅豆)等8菌株分別配對比較,並換算得各菌株間之相似率(similarity),用以鑑別分離株間之親緣關係,同一種類尾孢菌採集自不同寄主者,其相似率CcC與CcM為19%、PcC1與PcS為33%、PcC2與PcS為30%、PcA與PcS為18%、PcA與PcC1為40%、PcA與PcC244%;不同種類尾孢菌採集自相同寄主者,其相似率PcC1為CcC為20%、PcC2與CcC為15%;採集自花生與山珠豆之C. arachidicola與P. bradburyae與其他各分離株配對比較之相似率皆低於24%;自不同地區採集豇豆黴媒病之兩分離株PcC1與PcC2,其相似率達70%。 |
英文摘要 | The differences in isozyme polymorphisms among Cercospora arachidicola (CaP), C. canescens (CcC and CcM), Pseudocercospora bradburyae (PbC) and P. cruenta (PcC1, PcC2, PcS and PcA) were compared by using polyacrylamide gel electrophoresis techniques. Of 13 enzymes analyzed, isozyme polymorphisms were dectected for acid phosphatase, alkaline phosphatase, catalase, diaphorase, esterase, formate dehydrogenase, s-glucosidase, glutamate dehydrogenase, glutamate oxaloacetate transaminase, malate dehydrogenase, malate enzyme, peroxidase and shikimate dehydrogenase. The similarities were caculated by isozyme polymorphisms of these eight isolates in pair, in order to identify the relationship among these isolates. The same species of Cercospora spp. or Pseudocercospora spp. were collected from different hosts. The similarity of CcC and CcM is 19%, this of PcC1 and PcS is 33%, this of PcC2 and PcS is 30%, this of PcA and PcS is 18%, this of PcA and PcC1 is 40%, and this of PcA and PcC2 is 44%. The different species were collected from the same hosts. The similarity of PcC1 and CcC is 20%, this of PcC2 and CcC is 15%. The similarity of C. arachidicola and P. bradburyae collected from peanut and centrosema compared with other isolates is lower than 24%. The similarity of two isolates of PcC1 and PcC2 collected from different areas of cowpea is 70%. |
