中文摘要 | 傳統以形態特徵為依據的方法無法客觀鑑別冬蟲夏草屬(Cordyceps)真菌,因此本 實驗利用18S rRNA基因的限制□水解多型性圖譜(RFLP)來鑑別冬蟲夏草(Cordyceps sinensis (Berk.) Sacc.)與其它蟲草屬菌株。在以引子組NS1、NS4及NS3、NS6進行聚合□ 連鎖反應後,所得產物再分別經限制□Hae Ill、Hpa Il、Sau 3A、Rsa I及Cf0 I水解。結 果顯示冬蟲夏草(C. sinensis)的菌核與子座具有獨特的Cfo I限制□水解多型性圖譜,可用 來將冬蟲夏草(C. sinensis)標本與蟲草屬菌株作明確的區別。而利用此PCR-RFLP的結果將 有助於天然冬蟲夏草(C. sinensis)樣品的分類與鑑定。 |
英文摘要 | The identification and taxonomy of Cordyceps by the traditional method based upon morphological observation is often debatable. Restriction fragment length polymorphism (RFLP) of 18S rRNA gene was used to differentiate Cordyceps - sinensis (Berk.) Sacc. from other Cordyceps spp. in this study. Polymerase chain reaction (PCR) was performed using NS1, NS4 and NS3, NS6 as primers, the amplified PCR products were then digested by Hae III, Hpa II, Sau 3A, Rsa I and Cfo I, respectively. The polymorphism of the Cfo I-digested fragments was found to be specific to the scierotium and stroma of C. sinensis and could be differentiated from other Cordyceps spp. The results indicated that PCR-RFLP might be a useful approach for the identification and taxonomy of C. sinensis specimens collected from fields. |
