中文摘要 | 基隆山藥塊莖經丙酮乾燥處理成粉末後,再經系列步驟純化多酚氧化,純化之多酚氧化以L-3,4-dihydroxyphenylalanine(L-DOPA)及兒茶酚(pyrocatechol)為基質測定其力學機制。以L-DOPA為基質,此多酚氧化顯示Michaelis-Menten型動力學,Km值為190mM,然以兒茶酚為基質,則顯示歇斯底里型動力學,由動力學分析得知此種歇斯底里型反應屬結合基-誘導同分異構化歇斯底里型機制。 |
英文摘要 | Polyphenol oxidase was purified frm acetone powder of Keelong yam tubers through a series of sequential purification steps. The kinetic mechanism of the purified enzyme was studied by using L-DOPA or pyrocatechol as a substrate. The enzyme exhibited the Michaelis-Menten kinetic with L-DOPA as the substrate. The Km value was 190mM. However, the enzyme showed hysteresis with pyrocat-echol as the substrate. Results of kinetic analysis suggested that the hysteresis reaction behavior of the enzyme involves the ligand-induced isomerization hysteresis mechanism. |
