中文摘要 | IL-1β和TNF-α皆為可促成鈣化組織吸收的炎性介質,它們與牙髓炎或牙根尖病 變等組織炎性反應關係密切。為了探討IL-1β與TNF-α對人類牙髓細胞DNA合成與 蛋白質合成的調控作用,本研究以不同濃度之IL-1β與TNF-α刺激培養中之人類牙髓 細胞,進而測定細胞的[�鯨]-thymidine incorporation 及由三氯醋酸所沉殿之蛋白質的[ �鯨] proline incorporation 分別檢視IL-1β和TNF-α對DNA合成與蛋白質合成的影 響。研究結果顯示IL-1β(>500 U/ml)與TNF-α(>1 U/ml)皆會刺激牙髓細胞合成 DNA,IL-1β濃度為500 U/ml時,DNA合成增加為1.3倍,TNF-α濃度為1∼ 500 U/ml時,DNA合成增加為1.2∼1.5倍。此外,IL-1β在濃度高於50 U/ml時, 會抑制蛋白質合成,而TNF-α在0.1∼500 U/ml之範圍內,則皆未表現促進或抑制的 效果,上述結果顯示IL-1β與TNF-α極可能在人類牙髓組織的癒合或炎性反應中扮演 重要的調控角色。 |
英文摘要 | Interleukin-1β (IL-1β)and tumor necrosis factor-α (TNF-α)are cytokines that are activated during various inflammatory processes. The purpose of this paper is to elucidate their roles in the pulp tissue inflammation. Effects of IL-1 β and TNF- α on DNA synthesis in cultured human dental pulp cells were investigated by measuring the [ �� H]-thymidine incorporation into cellular DNA. The effects of IL-1 β and TNF- α on protein synthesis were analyzed by [ �� H]-proline incorporation into trichloroacetic acid (TCA)-precipitated proteins. Results indicated that IL-1 β (>500 U/ml)and TNF- α (>1 U/ml) stimulated DNA synthesis in pulp cells. The rate of DNA synthesis increased 1.3-fold by stimulation with 500 U/ml IL-1 β. TNF- α also stimulated DNA synthesis by 1.2 ∼ 1.5-fold (p<0.05) at concentrations ranging from 1 to 500 U/ml. In addition, IL-1 β suppressed 23 ∼ 27% of protein synthesis in pulp cells at concentrations higher than 50 U/ml (p<0.05). TNF- α exerted neither stimulatory nor inhibitory effects on protein synthesis at concentrations ranging from 0.1 to 500 U/ml. These results show that IL-1 β and TNF- α might be important factors in the inflammatory responses and healing processes in human dental pulp. |
