中文摘要 | 在研究青光眼的機轉或是降眼壓藥物作用機轉時,若能建立類似人類青光眼變 化的動物模式則其研究結果較能預測在人體上真正的變化。由於Alpha-chymotrypsin後房 注射產生之高眼壓兔子常被用來當作模式動物,而其房水動力學變化未曾有文獻報告過, 故在本研究中利用Alpha-chymotrypsin注射入兔眼後房以研究其房水動力學變化,結果發 現兔眼經Alpha-chymotrypsin注射後眼壓即急劇升高,但在術後兩週內眼壓有較大之起伏 變化,自二個月後至二年之間,經Alpha-chymotrypsin誘發之青光眼,其眼壓(平均值 38.4mmHg)則皆維持在較正常眼(平均值18.3mmHg)為高之水平(P<0.01)。此外在高眼壓兔 眼有較大之前房體積、虹膜-睫狀體複合體有明顯充血現象及水晶體有高比例之脫位( 38%)情形。 利用此一模式研究不同體積之Balanced Salt Solution(BSS)瞬時注入前房後,等量 之體積是否產生相同之眼壓上升時,可以發現其再現性良好。若逐次微量的將BSS加入前 房中可以觀察到眼壓上升之現象:加入之體積愈多,眼壓上升之幅度愈大,但在高眼壓眼 需加入較正常眼為多之BSS體積始可達同幅度之眼壓上升。若以眼壓開始上升至完全回復 至基線(Baseline)的時間為過量前房體積之排除時間,則可以發現在正常眼所加入之BSS 體積量與排除時間之關係為拋物線形,而高眼壓眼由於額外加入之體積在前房會有逐漸蓄 積的現象,故基線不斷的升高,因此無法建立排除時間與加入體積之關係。 |
英文摘要 | When we study the mechanisms of glaucoma or antialaucoma agents, it is beneficial and helpful to have animal models that simulate the changes of human glaucoma. Alpha-chymotrysin-induced ocular hypertensive rabbit eyes ( AOHRE) were frequently used as an animal model in studying glaucoma, yet the changes of aqueous humor dynamics of such a model have never been described. In this study, we reported the aqueous humor dynamics in AOHRE. It was found that the intraocular pressure (IOP) of the rabbits dramatically increased immediately after the injection of alpha-chymotrypsin. Although there was variation in IOP levels within the first two weeks after alpha-chymotrypsin injection, yet from two months to two years, the AOHRE showed significantly elevated I0Ps (mean: 38.4 mmHg) than those of the normal controls (mean: 18. 3 mmHg) (p<0.01). In addition to that, the AOHRE had larger anterior chamber volume, obvious injection on the surface of iris-ciliary body complex and higher incidence of lens dislocation (38%). In studying the relationship between extra added volumes and I0Ps of AOHRE and normal controls, it was found that the reproducibility was good between instant anterior chamber injection of Balanced Salt Solution (BSS) and IOP changes. The larger the amount injected into the anterior chamber, the higher the IOP level would be. But in AOHRE, it needed more volumes of BSS than in normal controls to result in the same amplitude of IOP elevation. In addition, if we defined that the time needed for the extra-volume of BSS to be excreted as excretion time, it was found that the excretion profile for the normal controls would be a parabolic curve, while in AOHRE, due to gradual accumulation of BSS in the anterior chamber, there was no such relationship existed. |
