英文摘要 | The efficiency of nuclear transplantation depends mainly on the enucleated rate, activation of recipient oocyte and synchrony of cell cycle stage between nucleus and cytoplasm. The purpose of this study was to confirm the relationship of collection time with the oocyte chromosome position in mouse and rabbit. The position of oocyte chromosome can be divided into three regions according to the distance relatively to the site of first polar body (PBI). The distance of the first region starts from the oocytes membrane in touch with the PBI to the membrane of the opposite site. The results showed the proportion of the intact PBI morphological integrity and the chromosome located in region I of oocytes in mice which were collected in early stage (10~11 h after hCG injection) was significantly higher than those collected in mid (12~13 h after hCG injection) or late (14~15 h after hCG injection) stages [73.9% vs. 48.7% or 14.1% (P<0.001) for PBI morphological integrity; 66.1% vs. 45.2% or 33.2% (P<0.05) for oocyte chromosome located in I region]. But this situation was not found in rabbit. For the collection time at 12~13, 14~15 and 16~17 h after hCG injection, the change of the proportion of PBI morphological integrity (87.0%, 89.0% and 83.3%) and of the oocyte chromosome located in region I (93.1%, 87.1% and 93.4%) were not significantly different. It is concluded, therefore, that the timing for oocyte collection has a dramatical influence on the convenience and efficiency of the performance of enucleation. The best time window of oocyte collection is 10 to 11 h after hCG injection in mice, while it is 14 to 17 h after hCG injection in rabbits. |
