中文摘要 | 光分解作用時,水分子被分解成三部份即氫離子、電子對和氧氣。在植體內擔負 此一重任的是水分解蛋白複合體, 它是由三個核基因產物 OEC16、OEC23 和 OEC33 蛋白共 同組成的。由於生化或生理研究皆未能完全解開水分解蛋白複合體三個蛋白的功用之謎,因 此利用轉殖植物帶有外來基因的特質, 將 OEC16 蛋白基因 (oee3) 殖入大豆體胚,應該對 明瞭該蛋白質在光分解作用所扮演之角色具有重要意義。 為了達成基因轉殖的目的, 建構 OEC16 蛋白基因表現組是首要工作。將由玉米分離的水分解蛋白複合體 16kD 蛋白基因先分 別以順向及反向選殖入 pRT106 或 pRT105 質體, 形成具有 35S 啟動子及 poly A 尾端的 pOE17S106 及 pOE17AS105 質體。考慮到大豆體胚轉殖後基因轉殖效率偵側與轉殖體篩檢的 需要,再將 OEC16 基因表現組重新選殖到 pCAMBIA1201 質體上,使轉殖用質體同時具有抗 生素 Hygromycin B(hpt Ⅱ )、GUS(gusA) 及 oee3 三個基因。 |
英文摘要 | In photolysis, water molecule was spliced into protons, electrons, and oxygen molecules. Oxygen-evolving complex (OEC) is responsible for this reaction during photosynthesis. The OEC complex is composed of three nuclear-encoded gene products, named OEC16, OEC23, and OEC33. Functional studies for individual protein of the complex by either physiological or biochemical methods are not very successful. Thus, transgenic plant studies on soybean which contain the OEC16 gene will greatly helpful in understanding it role in photolysis. In order to accomplish this goal, the maize OEC16 gene (oee3) was fused either sense (pOE17S106) or anti-sense (pOE17AS105) orientation to the CaMV 35S promoter at the 5'-end and its corresponding polyadenylation [poly(A)] signal at the 3'-end then the OEC16 gene expression cassette was subcloned into the pCAMBIA1201 plasmid which contains the Hygromycin �� (hpt Ⅱ )and the GUS (gusA) reporter gene for transformation detection and transformant selection. |
