中文摘要 | 臨床上常需要用到生物組織材料來修補或替代病變的組織或器官,這些材料在植 入人體前皆必須經過交聯理。然而目前臨床上用來交聯處理生物組織的交聯劑毒性都相當的 高,使得這些生物組織材料在臨床使用了許多的問題。為了克服這些問題,我們著手從中藥 桅子的果實裡萃取出一種毒性極低的天然交聯劑 -genipin。 本研究主要的目的是在探討 genipin 的萃取與純化及其對生物組織的交聯性質與其水溶液的驗結果顯示,我們從梔子果 實裡萃取純化出來的 genipin 純度可高達 98 的以上。 另外交聯指數、變性溫度與機械強 度的測量結果, 也証實了 genipin 對生物組織有相當好的交聯效果。 然而我們發現 genipin 在不同溫度及 pH 值的水溶液裡均有降解的現象, 其中又以高溫及鹼性的環境對 genipin 的降解影響較為嚴重, 因此 genipin 交聯生物組織較適合在室溫與中性的條件下 進行。在本研究裡,我們也以不同的化學試劑及 FTI 測試結果,試圖來解釋 genipin 與生 物組織交聯反應的機制。 |
英文摘要 | Bioprostheses derived from biological tissues can be used to repair or replace defect tissues or organs in the clinical applications. These biological tissues must be chemically modified by a crosslinking reagent before they can be implanted in humans. Various synthetic crosslinking reagents have been used in chemically modifying biological tissues. However, these synthetic crosslinking reagents are all highly cytotoxic. It is therefore desirable to provide a crosslinking reagent suitable for use in biomedical applications which is of low cytotoxicity and forms stable and biocompatible crosslinked products. In the study, we successfully purified a naturally occurring crosslinking reagent (genipin) from gardenia fruits, a traditional Chinese medicine. The cytotoxicity ofgenipin has been previously shown significantly lower than those of the crosslinking reagents commonly used clinically. The purity ofgenipin obtained was approximately 98%. It was shown that genipin is an effective crosslinking reagent for biological tissue fixation. Additionally, it was found that aqueous genipin was not stable in high temperature and basic environments. Therefore, it is suggested that fixation of biological tissue by genipin be conducted at room temperature and neutral conditions. The presumable reaction mechanism ofgenipin with biological tissue was also discussed in the study. |
