中文摘要 | 非構造蛋白質(nsPl)是辛德比斯病毒的轉甲基酵素,專門修飾病毒基因與副基因,是本病毒在複製時極為重要的蛋白質。為分析它的合成、安定性與酵素活性,我們利用抗 -nsPl 多株體免疫沉殿[□ S-Met] 標示的辛德比斯病毒感染蚊細胞,結果發現細胞在感染病毒後2小時開始合成nsP1產量最高點出現在感染後24小時;其合成量與細胞的培養溫度有關。28℃時nsPl,並持續至感染後4小時,而後逐漸降低;37℃時,nsPl則是自合成後,產量便呈持平狀態,而且持續24小時不變;若將感染細胞培養於34.5℃,它的產量與37℃時相似。此外nsP1極為穩定,例如蚊細胞在感染後5小時所合成的nsPl可保有近一天的安定性。我們以S-adenosylmenthionine為受質測定nsPl 的活性,發現不論蚊細胞培養在 28℃、34.5℃或37℃,nsPl的酵素活性都可在病毒感染後6小時測得,10小時的活性最高,且持續至24小時。 |
英文摘要 | To investigate the synthesis and turnover of the nonstructure protein, nsPl, in Aedes albopictus cells, we labeled infected cells with [□S]-methionine and immunoprecipitated nsPl with a polyclonal monospecific rabbit antibody. Synthesis of nsPl in nosquito cells could be detected 2 hr after infection and continued as long as 24 hr post-infection, regardless whether the infected cells were maintained at 28℃, 34.5℃, or 37℃. Wherease the time pattern of nsPl synthesis varied with temperature. Nonstructural protein 1 synthesis at 28℃ was maximal at 4 hr, then decreased. At 37℃ synthesis reached a high level at 2 hr and remained constant for 24 hr. Pulse-chase experiments showed that nsPl in mosquito cells, whether made as early as 5 hr post-infection or as late as 24 hr, were stable during that time, the longest period tested. Enzyme activity reached a maximum at 10 hr and remained almost the same level until 24 hr. The yield of Sindbis virus from mosquito cells was higher at 34.5℃ and 37℃ than at 28℃. Methyltransferase activity was needed for modification of positive-strand genomic and subgenomic RNAs. The activity of methyltransferease observed until late in the replication cycle probably accounted for both the continued synthesis and minimal turnover of nsPl. |
