中文摘要 | 本文探討臺桑二號(Morus alba L. var. acidosa G.cv. Taisang 2)桑葉甲醇萃取物之抗氧化性及其作用機制。以亞麻油酸為氧化受質,測定其抗氧化性,結果顯示桑葉甲醇萃取物具有與丁基羥基甲氧苯(BHA)相似的抗氧化性。在清除自由基及活性氧方面,當桑葉甲醇萃取物添加劑量在50 μg時對a,a-diphenyl-β-picryl-hydrazyl自由基具有83.2%之清除能力,顯示其具有提供氫原子以終止脂質氧化連鎖化應之一級抗氧化劑的作用型式。當桑葉甲醇萃取物添加劑量在100μg時,其對過氧化氫及超氧陰離子(superoxide anion)可達92.8%及57.8%的最大清除能力。而以電子自旋共振光譜儀(EPR)測定其對氫氧自由基(hydroxyl radical)的清除效應,發現隨著添加劑量的增加,其清除能力愈強,在10mg時可達90.2%的清除效果。因此桑葉甲醇萃取物亦是一種活性氧(reactive oxygen species)的清除劑,屬於二級抗氧化劑之作用型式。由本研究結果可知桑葉甲醇萃取物在抑制脂質氧化反應上同時扮演了一級及二級抗氧化劑之角色。 |
英文摘要 | The antioxidative activity and the mechanism of methanolic extracts of mulberry leaves (MEML) (Morus alba L. var. acidosa G. cv. Taisang 2) were investigated. The antioxidative activity of MEML on the peroxidation of linoleic acid was equal to that on the peroxidation of butylated hydroxyanisole. The scavenging effect of MEML on the a, a-diphenyl-β-picryl-hydrazy radical, was 83.2% at a dose of 50 μg, indicating that MEML has an effective hydrogen-donating activity and acts as a primary antioxidant on lipid peroxidation. MEML also showed a 92.8% and 57.8% scavenging effect on hydrogen peroxide and superoxide anion, respectively, at a dose of 100 μg. MEML exhibited a marked scavenging effect on the hydroxyl radical with 90.2% at a dose of 10 mg as measured by EPR. These results indicated that MEML was a scavenger for reactive oxygen species, and that it acted as a secondary antioxidant. The overall antioxidant effect of MEML on lipid peroxidation might be attributable to its properties as a primary and as a secondary antioxidant. |
