中文摘要 | 食品中葉酸的存在型式相當多,而微生物分析法可對所有不同型態葉酸化合物進行檢測,而且偵測極限亦可達到10-9g,使得微生物法仍被廣泛用以分析食品中之總葉酸含量。文獻中對於Lactobacillus casei(ATCC 7469)檢測葉酸之探討頗多,但對於Enterococcus hirae (atcc 8043)應用則討論較少,對於以E. hirae分析食品中總葉酸含量的文獻報告更是少見。由於食品中的葉酸大都與蛋白質、多醣體鍵結,加上其含有多個麩胺酸分子,因此在分析時萃取過程格外重要。在本研究中利用二階段萃取方式檢測食品中總葉酸含量,並比較上述二菌株對食品中葉酸之分析能力。結果顯示,樣品必須添加酵素進行預處理,才不致於低估葉酸總量。在使用豬腎酵素進行切割作用(deconjugation)時,反應的pH值宜控制在4.5左右。利用3ml經過純化的酵素液反應1小時,即可使食品中之葉酸全部被釋放。利用E. hirae與L. casei對於食品總葉酸含量之分析結果顯示,E. hirae可得較高之分析值。 |
英文摘要 | Folic acid presents in various forms in food systems. The microbiological method has been proved to be one of the most effective ways to analyze the total folic acid content in foods. The detection limit of this method can be as low as pictogram (10-9g). Most researches reported in the literature have focued on Lactobacillus casei (ATCC 7469); of Enterococcus hirae (ATCC 8043) has scarcely been used for this task. Due to the complications of folic acid that it is usually chemically bound to proteins and polysaccharides, and that it contains different numbers of glutamic acid molecules, the extraction procedure prior to sample analysis is crucial In this study, two-step extraction was used for the sample preparation. Two different species of microorganisms (E. hirae and L. casei) were also examined. Results showed that enzymatic pretreatrment of the sample was necessary to improve the accuracy of this method. Enzyme obtained from hog kidney was used for the deconjugation of the sample. Folic acid can be totally released from the food matrix by using 3 ml of the purified enzyme solution. The reaction was carried out at about pH 4.5 and continued for a period above 1 hr. For this microbiological method of analysis, it was found that the use of E. hirae could result in higher analytical values than could be obtained using L. casei. |
