中文摘要 | 水溶性多醣類在生理效用之價值已被廣泛研究,而一些菇類子實體中亦可萃取出水溶性多醣類。本研究使用兩種猴頭菇菌株(Hericium erinaceus及Hericium laciniatum)探討以液體浸漬振盪培養方式,產製水溶性多醣類之最適當培養基組成、培養基起始酸鹼值、接種量及溫度,實驗結果利用統計學上之Duncan multiple方法進一步分析加以確認。醱酵期間菌絲體的增加對水溶性多醣類產量的增加具正面效果,而培養過程酸鹼值的過度變化則不利於水溶性多醣類的生成。培養基組成以含有機物成份之培養基在水溶性多醣類產量上多於單純氮源組成之培養基,且碳氮比以40及50為宜(p<0.05);而培養溫度在24℃時,於第25天後,水溶性多醣類產量明顯增加,至第30天後之增加量,與其他三種培養溫度呈現明顯差異(p<0.05)。培養基起始酸鹼值在5.5時,於發酵之第20天後,雖然培養基之酸鹼值明顯下降,但大致上其水溶性多醣類產量之累積上升情形為四種起始酸鹼值中最高者(p<0.05)。接種量以10%(v/v)在水溶性多醣類產量之效果最好(p<0.05)。兩種試驗菌在幾種特定培養條件下,Hericium erinaceus之水溶性多醣類產量均優於Hericium Laciniatum。 |
英文摘要 | The physiological effects of water-soluble polysaccharides have been studied extensively. A few of them may be extracted from the fruiting body of some mushrooms. In this study, Hericium erinaceus and Hericium laciniatum were cultivated in submerged shaking culture to investigate the optimal composition of media, initial pH value, incubation temperature, and inocula to produce water-soluble polysaccharides. The Duncan multiple method was employed to confirm the data The yields of water-soluble polysaccharides were closely related to the increment of the mycelial weight during fermentation. Drastic change in the pH value was unfavorable to the production of water-soluble polysaccharides. As to the effects of medium composition, it was found that yields under complex media were higher than those of simple nitrogen sources. In addition, the media with C/N ratios of 40 and 50 also produced better yields (p<0.05). Also, the production of water-soluble polysaccharides was considerably increased when incubation lasted for 25 days at 24℃, and the increment of the accumulated yields showed a significant difference (p<0.05) in comparison with the other three kinds of cultivation temperatures after 30 days. Although the pH value of the fermented broth obviously dropped after 20 days at pH 5.5, it was the value under which the highest yield was obtained among the four initially tested pH values (p<0.05). It would be more appropriate with 10% (v/v) inoculum for favorable production of water-soluble polysaccharides (p<0.05). Under selected cultural conditions, Hericium erinaceus had higher yield of water-soluble polysaccharides than did Hericium laciniatum. |
