英文摘要 | The protein compositions and their molecular sizes are quite similar between goat's milk and cow's milk. However, the native charges of their respective protein are variant in each other. Urea polyacrylamide gel (urea-PAGE) electrophoresis revealed that most of goat's milk proteins showed less charges than cow's milk protein exceptβ-casein. The concentration of αs1-casein was high in cow's milk and it moved fast at electrophoresis and strong band after gel staining. And fortunately goat's milk proteins did not show the same band at the position of αs1-casein of cow's milk after alkaline electrophoresis. Thereforeαs1-casein of cow's milk became a marker for detection of cow's milk in goat milk. Optimal conditions wre established for alkaline urea PAGE using 1.5 mm thick gel plate and 15μl sampling volume. The profiles of those milk proteins showed that casein of cow's milk and goat's milk resolved extremely well. The sensitivity of detection was so high that as little as 0.5% (v/v) of cow's milk in goat's milk could be detected. |
