英文摘要 | Cardiovascular cells interact with one another via gap junctions, cell membrane structures which consist of clusters of channels permitting direct intercellular communication. The component proteins of these junctions are termed connexins, different members of which contribute to the unique properties of the individual channels. As gap junctional channels allow passage of electrical and metabolic signals between adjacent cells, a variety of biological activities depend on gap junctions. In heart, cardiomyocytes are extensively interconnected by gap junctions and act as a functional syncytium. The coordinated contraction of cardiac muscle in response to pacemaker's command enable effective pumping. In vascular wall, gap junctions participate in the repairing mechanism and the modulation of vascular tone, which is pivotal to the maintenance of homeostasis of circulation. As gap junctions are involved in major cardiovascular functions, the relationship between gap junctions and cardiovascular diseases have been investigated and emphasized. Evidences from several lines of studies show that diseases can alter gap junction distribution and connexin expression. On the other hand, abnormality of connexin can initiate disease processes. Such connection have been demonstrated in different categories of cardiovascular diseases, for example, congenital heart disease, ischemic heart disease, hypertrophic cardiomyopathy, cardiac arrhythmia, as well as vascular pathologies of hypertension, atherosclerosis, and after balloon catheter injury. As the role of gap junctions in healthy and diseased states becomes clarified, efforts from these researches are sure to pave a way for novel intervention in cardiovascular diseases in the near future.
