中文摘要 | 目的:探討雄性素接受體的多麩胺數目的多寡是否與精子的製造不良有關。 方法:我們一共收集了190位不明原因的男性不孕症病人及101位有生育能力的健康男 性的血液樣本。由周邊血液的淋巴球萃取出DNA,經由PCR反應放大含有CAG重覆的DNA片 段。以Genescan分析儀將CAG的重覆數目計算出來。為了証實Genescan分析儀的準確性, 我們將部分的樣本用序列分析測量其CAG重覆的數目。 結果:在男性不孕症患者及正常男性,其雄性素接受體多麩胺數目的中間值分別為24及 23。當多麩胺的數目超過28以上時,其精子製造不良的機會會有2倍以上的增加。相反的, 若多麩胺的數目小於22時,精子製造不良的機會將大幅減少。 結論:雄性素接受體多麩胺數目的多寡與缺損性精子的機率有關。 |
英文摘要 | Objective: To investigate whether the lengths of polyglutamine tracts in the androgen receptor (AR) are related to defective sperm production. Methods: We examined the lengths of the polymorphic CAG repeat segment of the AR gene in 190 individuals with idiopathic male infertility due to defective spermatogenesis, and compared them to 101 healthy fertile male control subjects. Genomic DNA was extracted from peripheral lymphocytes of the patients with defective spermatogenesis and the fertile controls. The DNA fragment containing the CAG repeats was specifically amplified and its size determined with an automated Genescan analysis with internal size standards for each sample. Accuracy of size assignations was confirmed by sequencing some GAG repeat fragments. Results: The median glutamine numbers for subjects with male infertility and controls were 24 and 23, respectively. Subjects with long polyglutamine tracts had a 2-fold increased risk of defective spermatogenesis. The risk of having severe oligospermia increased from 1.75-fold in patients with 26 or more glutamine repeats, to 3-fold in those with 28 or more repeats. Short polyglutamine tracts of 22 glutamines or less were associated with reduced risk of defective spermatogenesis. Conclusions: The data indicate an association exists between the length of the polyglutamine tract in the AR and the risk of impaired spermatogenesis. |
