中文摘要 | Corynebacterium glutamicum在8%氯化鈉發酵培養基中,經72小時發酵,脯胺酸 產量高於無添加鹽對照組約八倍。氯化鈉是優於硫酸銨的氮源,適當濃度是2%;自7%氯 化鈉和2%氯化氨發酵培養基中,葡萄糖濃度以3-4%為宜,超過4%則不利於脯胺酸的合成; 生物素濃度應控制在20-40μg/L。由5L小型發酵槽批式發酵結果得知,在含高濃度氯化氨 培養基中,脯胺酸的產生與菌體生長同步,麩胺酸的產生在菌體進入對數生長初期,即開始 衰退,而此刻正是脯胺酸開始快速增加的時後。這兩種胺基酸生合成的消長,意味著脯胺酸 合成的增加,可能是由相容性溶質麩胺酸轉變而成。 |
英文摘要 | Proline production of Corynebacterium glutamicum in media containing 8% NaCl was eight times higher than that of the control. The optimal concentrations of NH Cl, glucose, and biotin for production of proline in the media containing 7% NaCl were 2%, 3-4%, and 20-40μg/L, respectively. According to a batch fermentation performed in a 5-liter fermentor, the proline production was associated with proliferation of bacterial cell. The concentration of proline increased in the culture, on the contrary, the amount of glutamate reduced. This result implied that formation of extra proline in the media containing high concentration of NaCl might be due to the conversion of glutamate. |
