中文摘要 | 對大部分的粒線體來說,ADP 依賴型呼吸作用是一種普遍的現象。然而,本研究 發現粒線體在沒有 EGTA 存在的情況下, 不具有 ADP 依賴型呼吸作用, 但是在 15 μ MEGTA 存在的情況下,粒線體恢復此呼吸作用,文中亦討論幾種可能引起此種異常現象的原 因。 本研究亦評估重金屬對剛分離吳郭魚 (Oreochromis mossambicus) 肝臟粒線體的毒性 ,其毒性原理在於重金屬干擾粒線體呼吸酵素活性的能力。 重金屬對粒線體狀態 3 的呼吸 速率具有抑制作用, 然而它對狀態 4 的呼吸速率卻有輕微的刺激作用,而且四種重金屬( 鎘, 汞,鉛和砷)抑制粒線體狀態 3 呼吸速率一半的濃度分別為 0.4 ppm, 0.8 ppm, 1 ppm 和 5.3 ppm,對粒線體的毒性順序為鎘>汞>鉛>砷。同時,本研究的結果亦顯示汞金 屬的毒性形式以游離態為主,而非複合型 (complexed form)。 |
英文摘要 | For most mitochondria, ADP-dependent respiration is a common phenomenon. However, in the present study, no ADP-dependent respiration was observed in tilapia mitochondria when EGTA was absent in the assay but the respiration was recovered after the addition of EGTA to a final concentration of 15 μM. The possible reasons for this unusual phenomenon were discussed. This study also evaluated the toxicities of heavy-metal ions on the respiration of mitochondria freshly isolated from the liver of tilapia (Oreochromis mossambicus). This method measured the ability of heavy metals interfering the mitochondrial respiratory enzymes. Mitochondrial respiration in state Ⅲ was decreased by increasing the concentration of heavy metal ions, while the respiration rate in state Ⅳ was slightly increased. The order of toxicity of four heavy metals on mitochondrial respiration rate in state Ⅲ was Cd�ョ烺g�ョ烳b�ョ淲s�� in the presence of 15μ M EGTA. The results of this study also showed that the free form instead of the complexed form of mercury ions exerted the toxicity. |
