| 中文摘要 | 停經後的婦女容易造成骨質疏鬆症,這些現象一般認為與體內鈣的代謝有密切的關係,以中醫基礎理論立場而言,是由於屬於腎陰不足,腎精虧損所致,所以本研究的目的是探討河車大造丸對切除兩側卵巢後老鼠鈣的效用,更進一步尋求新的方法來治療停經後之骨質疏鬆症。將35隻重約200克、90天齡之雌性老鼠,隨機將它們分成每7隻一組如下:1)假性卵巢切除組(SH):將老鼠兩側的卵巢取出放置於腹腔外然後再放,但不切除;2)兩側卵巢切除組(OV):將老鼠兩側的卵巢切除;3)兩側的卵巢切除加河車大造丸組(OV+D)將老鼠兩側的卵巢切除28天後投予河車大造丸至56天犧牲為止;4)假性卵巢切除加河車大造丸組(SH+D):同假性卵巢切除組,但投予河車大造丸方法如同兩側的卵巢切除加河車大造丸組;5)控制組(Control):不施行任何手術或投予任何藥物。我們觀察它們的體重變化,血清中鈣、副甲狀腺賀爾蒙和鹼性磷酸�t以及尿中Pyrilinks-D濃度。結果發現河車大造丸可減少兩側卵巢切除老鼠的體重、鹼性磷酸�t和尿中Pyrilinks-D增加,提高鈣與副甲狀腺素的血清濃度。根據這些結果我們認為河車大造丸可以調節兩側卵巢切除後老鼠鈣的代謝,如此推論河車大造丸對停經後所造成的骨質疏鬆症可能有治療的效果。 |
| 英文摘要 | The post-menopause osteoporosis is due to the remodeling of the bone and release of the calcium from the bone. According traditional Chinese Medical theory, osteoporosis results from kidney-yin vacuity and insufficiency of kidney-essence. Ho-Tr-Da-Za-Wa has the effect of supplementing the kidney, replenishing essence and marrow, strengthening the bone, enriching yin and clearing heat. In order to study an effective method to improve the metabolism of calcium for treating post-menopause osteoporosis, we used Ho-Tr-Da-Za-Wa to ellucidate its effect on calcium metabolism of rats with bilateral ovariectomy. A total of 35 female Sprague-Dawly (SD) rats (200g in body weight, 90 days in age) were divided into 5 groups of 7 rats as follows: 1) Sham ovariectomy group (SH): their bilateral ovaries were pulled to the outside of the body and recovered immediately. No ovariectomy was performed; 2) Ovariectomy group (OV): bilateral ovariectomy were done; 3) Ovariectomy +Ho-Tr-Da-Za-Wa group (OV+D): using Ho-Tr-Da-Za-Wa (100mg/ml/100g) orally from 28�裨o 56�蜿ay after bilateral ovariectomy were performed; 4) Sham ovariectomy +Ho-Tr-Da-Za-Wa group (SH+D): the procedure is the same as the SH group except using Ho-Tr-Da-Za-Wa (100mg/ml/100g) orally from 28�裨o 56�蜿ay after operation was performed; 5) Control group (Control): no operation was performed and no drugs were administered. We measured their body weight changes and also measured serum calcium, parathyroid hormone (PTH), alkaline phosphatase(ALP), and urine deoxypyridinoline(Pyr-D) and D-pyr concentration at 56�蜿ays after bilateral ovariectomy. Our results indicated that Ho-Tr-Da-Za-Wa decreased the increase in body weight, urine Pyr-D and ALP concentration, and increased the decrease of serum concentration of Ca and PTH, which resulted from rats with bilateral ovariectomy. Our conclusion are that Ho-Tr-Da-Za-Wa has a regulatory effect of calcium metabolism to rats with bilateral ovariectomy, suggesting that this formula may be used to osteoporosis resulting from menopause or bilateral ovariectomy in human. |
