中文摘要 | 本研究之主要目的在於測定及比較甜楓種子苗、組織培養苗,以及兩種轉殖外源甲基轉移脢(O-methyltransferase; OMT)核酸之轉殖苗木的生長表現及生理特性。測定及估算的項目,包括苗木高生長、地際直徑、淨光合速率、葉綠素濃度、葉部全氮量、苗木各部乾重、苗木總乾重、葉綠素a/b、梢根比(S/R ratio)和苗木品質指數。測定的對象為一個半月生的試管苗及移植到溫室4、6和8個月生的苗木。結果顯示,各項生理測定值與苗木品質估算值在不同處理苗木間之差異顯著性呈現不規則的變化,但是這些差異隨著苗齡的增加有逐漸減緩的趨勢,亦即8個月生的基因轉殖苗與種子苗及組織培養苗在生長與生理上已頗為近似,可以如同種子苗出栽造林。至於依據各項性狀數值之大小所排定的生長優劣情形而言,移植後8個月生的轉殖顫楊OMT cDNA正轉式(sense construct)之苗木在14項的測定與估算值中,有11項領先其他三種苗木,此一現象指出,正轉式外源OMT核酸之表現具有促進甜楓苗木生長與生理性狀之效應。 |
英文摘要 | The objectives of this study were to determine growth performance and physiological characteristics of sweetgum seedlings, tissue-cultured plantlets, and 2 types of transgenic plants carrying the foreign O-methyltransferase (OMT) gene. Growth traits and physiological characteristics including height, diameter at root collar, net photosynthesis, concentrations of chlorophyll a and b, total nitrogen in leaves, and dry weights of leaves, stems, and roots were measured for various plants. The chlorophyll a/b ratio, shoot and root (S/R) ratio, and Dickson index for plant quality were calculated. Plants were measured at 1.5 mo old in vitro, and at 4, 6, and 8 mo old after transplanting to the greenhouse. The results show that significant differences in physiological characteristics and quality parameters varied irregularly at different stages of plant development. However, differences were reduced as plant ages increased. The 8-mo-old transgenic plants carrying the full length of aspen OMT cDNA in sense construct led the other types of plants in 11 of 14 characteristics by ranking the measured or calculated data. This phenomenon indicated that the expression of foreign OMT cDNA in sense construct might stimulate growth traits and physiological characteristics of sweetgum plants. |
