中文摘要 | 本研究之目的在當試開發出一個簡單和快速檢測飼料中clenbuterol之方法。先以 市售商業化檢測;β -agonist 之酵素檢驗套組進行樣品篩選工作,擬陽性樣品再以高效液 相層析法搭配紫外光檢測器及電化學檢測器進行檢測。結果顯示樣品僅須經簡單地抽提後, 通過 Extrelut 20 管柱即可進行分析。 利用 Cosmosil 5C18-MS 分離管,以 25 %氰甲烷 溶液加 0.01M 三水醋酸鈉緩衝液 (利用醋酸調 pH 至 3.5) 為移動相, 於紫外光檢測波長 244nm 和電化學檢測 +1250mV 下進行分析,可成功地自飼料中檢測出 clenbuterol, 並且 平均回收率可達 79.4 %。 |
英文摘要 | The aim of this study is to develop a simple, rapid, and reliable method for determination of clenbuterol in feeds. Samples were performed by using enzymelinked immunosorbent assay (ELISA) as a screening method. Positive samples were confirmed by high performance liquid chromatography (HPLC) with series of ultraviolet and electrochemical detection.Clenbuterol in the sample was extracted with water (pH 3.5), followed by an Extrelut 20 columncleanup and HPLC as the means of measurement. Analy sis was carried out on a Cosmosil5C18-MS column (Nacalai, Japan) with acetonitrile- 0.01M sodium acetate buffer (25:75 v/v, pH3.5 adjusted with acetic acid ) as the mobile phase, and was detected both by UV detector (at244nm) and electrochemical detector (at + 1250 mV). The results revealed that clenbuterol in.feeds was detected in 7.9 min, and the average recovery was 79.4%. |
