中文摘要 | 年老之哺乳動物,神經系統的退行性變化已很清楚, 如中樞神經系統之大腦血管 旁神經膠纖維增生及基底膜厚度增加,周邊神經系統之交感神經節微血管數目減少及血管腔 總面積減少等變化,但在副交感神經系統組織的血管情形則不清楚。因此,本研究以不同年 齡群之大白鼠之心臟神經節進行定性與定量形態測量分析,以了解老化對副交感神經系統組 織中,微血管構造所造成的影響。 大白鼠於固定後,心臟之心房後壁與大血管取下,經鋨酸再固定,酒精脫水,塑膠包埋後, 切成厚片及薄片,經染色後,分別於光學顯微鏡與穿透式電子顯微鏡下觀察,並將電子顯微 鏡照得之血管影像,經由描繪器連接電腦測量分析。 光學顯微鏡觀察到大白鼠之心臟神經節可分為三種,第一種主要為主神經細胞構成,第二種 為主神經細胞與少量之含顆粒小細胞構成,第三種以含顆粒小細胞為主要構成。而在不同種 類之神經節,其血管之分佈亦呈現局部性之差異,而以在第三種心臟神經節之血管數目最多 ;此種血管分佈狀況與血管數目並未在年老老鼠呈顯著的變化。 電子顯微鏡觀察到大白鼠之心臟神經節之微血管有兩種型,連續型與篩洞型,而以連續型最 常見。在年老之大白鼠亦出現同樣的情形。血管形態測量經過電腦分析顯示在老化過程中, 微血管腔,週長及面積,以及內皮細胞之面積有增加,但無統計學上顯著之差異。 本研究顯示老化過程中,副交感神經節的微血管循環的變化並不顯著。 |
英文摘要 | It is well estamlished that regressive changes of the nervous system are found in aging mammals, for example, the proliferation of glia around cerebral vessels and an increment in basement membrane thickness can be found in the central nervous system. On the other hand, in the sympathetic nervous system, the number and total area of capillaries are reduced in the aging processes. However, the age-related changes of the capillary in parasympathetic gandlia are still unclear. Therefore, the purpose of this report was to study the cardiac gandlia in case of different age of rats by using light and electron microscopes. After anesthesia and vascular perfusion, the hearts of 1-, 18- and 24-month -old Long-Evans rats were postfixed in osmium tetroxide, dehydrated, and embedded in Araldite 502. The sections were observed with the light- and electron microscopes. In the atrial wall, three types of cardiac ganglia were recognized. The type 1 ganglia were characterized by the presence of principal neurons; the type 2 ganglia consisted of principal neurons and a small number of small granular cells; the type 3 ganglia were characterized by the presence of small granular cells and the absence of principal neurons. In different types of ganglia, the distribution pattern and the number of blood vessels showed regional variations. The type 3 ganglia consisted of abundant blood vessels which intermingled with small granular cells and formed a glumerular structure. However, during the aging process, the distribution pattern and the number of blood vessels did not show significant changes. The ultrastructural study showed numerous continuous capillaries and a few fenestrated capillaries distributed in the cardiac ganglia. Most of the fenestrated capillaries were found close to small granular cells. The morphometric analysis showed that the diameter, perimeter, and area of capillaries, and the size of endothelial cells increased in the cardiac ganglia of aged rats. However, the difference was not showed to be statistically significant. It is suggested that the aging process in rats did not caused significant changes in the microvasculature of the ganglia in the parasympathetic nervous system. |
