中文摘要 | 本研究經由菌種篩選及化學突變法改良Monascus sp.之Monacolin K 產量,以期獲 得一高產菌株。首先,由15株Monascus spp.篩選出Monascus pilosus NCHU 527,其monacolin K的產量較高,在平板培養基中培養所產生的孢子量也較多。經由培養基組成及培養溫度等 之探討,設計了命名為PMPA之平板培養基,而培養溫度則以28℃較佳。利用此一條件來 進行M. pilosus NCHU 527之平板培養,所得孢子量比使用傳統的PDA培養基約可提升一 倍。隨後,利用N-methyl-N'-nitro-N-nitrosoguanidine化學突變劑對M. pilosus NCHU 527進 行突變處理,並以含抑菌劑之平板培養基來篩選高產突變株。最後,由含econazole的平板 培養中篩選出一株突變株:M. pilosus NCHU M-35,在LY培養基中培養7天後,其monacolin K 產量達208mg/L,為野生菌株的10倍以上。 |
英文摘要 | In this study, monacolin K-hyperproducing strains of Monascus sp. were selected by means of chemical mutagenesis and screening. Experimental results indicated that Monascus pilosus NCHU 527 was relatively high in monacolin K production and spore formation among the fifteen different strains of Monascus spp. tested. Spore formation could be increased approximately one-fold by cultivating this culture in PMPA medium at 28℃. In addition, M. pilosus NCHU 527 was bred by treating its spores with N-methyl-N'-nitro-N-nitrosoguanidine and screening the mutants, which were able to grow in an agar medium containing drugs. An econazole-resistant mutant, M. pilosus NCHU M-35, was obtained. NCHU M-35 had a 10-fold increase in monacolin k production compared to that of wild-type strain NCHU 527. Furthermore, the maximal titer, 208 mg/L, could be achieved within 7 days in LY medium. |
