中文摘要 | 毛細管電永(CE)是最近新發展出來的分析工具,具有高效、快 速、選擇性佳的特性。為解決高效能液相層析(HPLC)在中藥高極性 成分分析上的困難,本研究乃以毛細管電泳技術定量含黃連黃柏的 中藥製劑,並用毛細管電泳及高效液相層析儀分析疏肝湯,效果良 好。 用50%的0.2M醋酸鈉溶液和50%的氰甲烷可在八分鐘內分別 分析二十三種含黃連黃柏製劑之coptisine 、berberine和 palmatine,其回收率在98.0-101.9%之間,相對標準偏差是0.96- 1.50%(intraday)和2.22-2.68%(interday)。 由於毛細管電泳的高選擇性,藉著使用三種不同的分析條件可以 分析出疏肝湯中的十八個化合物。在MECC中可同時分析 paeoniflorine, benzoic acid, neohesperidin, hesperidin, naringin, ligustilide, butylidenephthalide, umbelliferone;在 CZE中分析dehydroevodiamine, coptisine, berberine, epiberberine, columbamine, jatrorrhizine, palmatine;而以逆電 滲流毛細管區帶電泳 (reversed electroosmotic CZE)分析nicotinic acid, caffeic acid和ferulic acid 三個成分。其分析時間分別為 MECC:40分鐘;standard CZE:30分鐘;reversed EOF CZE:11分 鐘。 利用分析吳茱萸黃連藥對之醋酸/醋酸鈉沖提系統,及新開發的 磷酸/磷酸鉀沖提系統來分析此方劑,高效能液相層析儀共計可定量 疏肝湯中二十四個成分,分析時間前者為60分鐘後者為100分鐘。 |
英文摘要 | Capillary electrophoresis (CE) is a recently developed analytical instrument which possess the features of high efficiency, short analysis time and high selectivity. In order to overcome the diffidulties of HPLC in analyzing high polar components of Chinese herbal drug, the techniques of capillary electrophoresis have been applied to accomplish the determination of berberine in 23 chinese berbal preparations. The contents of components in Shu-kan-tang has also been dtermined by CE and HPLC. A buffer solution of 0.2 sodium acetate solution- acetonitrile(1:1) was used to determine the contents of coptisine, berberine and palmatine in 23 Coptidis Rhizoma- and/or Phellodendri Cortex-containing Chinese medicinal preparations within 8 min. The recoveries of these three alkaloids were 98.0-101.9%. The relative standard deviations were 0.96-1.50% (intraday) and 2.22-2.68% (interday). Eighteen components in Shu-kan-tang were separated by combining three different analytical methods. The MECC method was applied to analyze paeoniflorine, benzoic acid, neohesperidin, hesperidin, naringin, ligustilide, butylidenephthalide and umbelliferone in 40 min; CZE technique was used to determine coptisine, dehydroevodiamine, berberine, epiberberine, columbamine, jatrorrhizine, palmatine in coptis in 30 min; and reversed EOF CZE was used to analyzed nicotinic acid, caffeic acid and ferulic acid in 11 min. Meanwhile, 13 compounds of its 24 components can be determined by the method for analyzing coptis-evodia herb couple, and 18 compounds of its 24 components can be assayed by a LC method using gradient solvent system that contained phosphate buffer. The running times were 60 min for the former and 100 mi. for the latter. |
