英文摘要 | Thickness of the cumulus cells around the oocyte is a morphologic indicator for selecting the cumulus oocyte complex (COC), but the viability of the enclosed cumulus cell is difficult to determine instantly. A newly developed fluorescent stain SYBR-14 which is not detrimental to cellular function was used for assessing the viability of the cumulus cell in combination with the fluorescent stain PI. Microscopic examination revealed that SYBR-14 stanied the living cumulus cell bright green and PI stained the dead cell bright red. Hence, the cell viability of the cumulus layers could be assessed by the percentage of the number of fluoresced green cells to the total number of fluoresced green and red cells. A total of 262 COCs and 564 COCs were collected at I~2 hours and 4~6 hours postmortem, respectively, and they were stained fluorescently. The narrowest diameter of COC and the cell viability assessment in the cumulus layer were determined simultaneously for each of COCS. The mean diameter of COCs was 242±37 μm (mean±standard deviation) and there were 88±13% of cells alive when COCs were collected at 1~2 hours postmortem. At 4-6 hours postmortem, the mean diameter of COCs was 209±24μm and there were 56±33% of cells alive. Percentage of COCs with 100% living cumulus cells to total number of COCs was 14.9%. It indicated that the survival rate of the cumulus cells around the oocyte was low as they were recovered from ovaries at 4~6 hours postmortem. Linear regression analysis showed a positive relationship between the thickness of the COC and the percentage of viable cells in cumulus layers around the oocyte with 0.58 and 0.27 of correlation coefficient (P<0.001) either at 1~2 hours or 4~6 hours postmortem. The larger the diameter of the COC, the more living cells in the cumulus layer. |
