中文摘要 | 體外培養時,廣東血線蟲的第三期幼蟲與青春期成蟲出現活躍的表面脫落現象,脫落物的蛋白質圖譜與脫落動力學俱經報告。青春期成蟲之脫落物已被證實具有抗原性而第三期幼蟲者則否。本研究中先將蟲體以植物凝集素-Con A 吸附,再經過氧化氫及其受質之標示後進行超微構造觀察。結果顯示蟲體上表皮外有一層形狀不定的醣類外被存在,或呈擴散狀、或呈大小懸殊之顆粒狀,然而後者並不具備單位膜三層次的特徵。顆粒物在外皮內分佈於不同部位,有些趨近上表皮,有些則與其融合。本研究基於對此線蟲超微構造的觀察進而推測形成其外被的醣蛋白在外皮內的可能輸送途徑與分泌過程。 |
英文摘要 | Active surface shedding phenomena were found in third stage larvae and juvenile worms of Angiostrongylus cantonensis in vitro. Profiles of released surface proteins and shedding kinetics were previously published. The antigenicity of proteins shed from juveniles, but not from third stage larvae, was already proved. Ultrastructures of the surface of juveniles were observed after binding with Con A lectin and labelling with peroxidase in the present study. An amorphous, sugar residue-containing surface coat was visualized outside the epicuticle. Both the diffused and the vesicular forms, with dramatically different sizes but without the typical trilaminate characteristics of plasma membranes as viewed by electron microscopy, were present all over the body surface. True vesicles were dispersed within the cuticle. Their approach to and fusion with the epicuticle could be observed. Based on these ultrastructural observations, speculations are made on the possible transcuticular transport of glycoproteins which probably produces the surface coat, and the possible secretion process by exocytosis. |
