中文摘要 | 目的:為了闡明在台灣的良性卵巢畸胎瘤的染色體組成及其細胞來源。 方法:我們從16個罹患良性卵巢畸胎廇病人取得其未稍血液及腫瘤組織。用原 位法培養腫瘤細胞,並以培養盒方法培養末梢淋巴細胞。染色體均以G 顯帶後用Wright染料來染色,以記錄色體之數目及構造。為探討畸胎 廇的細胞來源,我們比較腫瘤細胞及病人末梢淋巴細胞中染色體1,9, 16之長臂及染色體13,14,15,21,22,之短臂的異型及染色體質部位之 多形性,可推斷腫瘤細胞的染色體來自卵母細胞的那一分裂階段。 結果:除了一個腫瘤組織(6%)有不正常之染色體核型47,xx,+7之外,所有 的腫瘤組織與病人之末梢淋巴細胞一樣有正常之46,xx染色體組成。有 十一個腫瘤組織至少有一個異合子型異型染色質。有一位病人,其腫瘤組 織之8個多形性標記為同合子型,然而其末梢淋巴細胞於染色體14的多 形性短臂標記為異合子型。其餘四個病人,其腫瘤組織及末梢淋巴細胞於 8個多形性標記為同合子型。 結論:大多數之良性卵巢畸胎瘤有正常之染色組成。初級卵母細胞的第一次減 數分裂失敗似乎可以解釋大多數良性卵巢畸胎瘤之細胞來源。 |
英文摘要 | Objective:To elucidate the chromosomal composition and cellular origin of benign ovarian tera-tomas in Taiwanese patients. Methods: We obtained tumor tissues and peripheral blood from 16 patients with benign ovarian teratomas. An in-situ cell culture was set up for tumor tissues and a flask culture was set up for peripheral ripheral lymphocytes. Chromosomes were stained with Wright stain after Giemsa banding. Numerical and structural abnormalities of the chromosomes were recorded. Polymorphisms in the heterochromatin regions of the long arms of chromosomes 1,9, and 16 and of the short arms of chromosomes 13,14,15,21, and 22 were compared between the tumor cells and the host peripheral lymphocytes to determine the cellular origin of the tumor tissues. Results: Except in one (6%) tumor tissue with an abnormal karyotype of 47,xx,+7, all tumor samples had a normal chromosome complement of 46,xx as did all peripheral lymphocyte samples. Eleven tumor samples were heterozygous in at least one heterochromatin polymorphism. In one patient, the tumor tissue was homozygous at all 8 polymorphic markers and the peripheral lymphocytes were heterozygous at the polymorphic short arm marker of chromosome 14. In the remaining 4 patients, both the tumor tissues and peripheral lymphocytes were homozygous at all 8 polymorphic markers. Conclusion:Most benign ovarian teratomas had a normal chromosome complement. Failure in meiosis I of the primary oocytes seemed to account for the cellular origin of most cases of benign ovarian teratomas. |
