中文摘要 | 本試驗旨在探討豬隻在不同時間手術取胚對原核胚回收率之影響與不同相對離心 力對原核顯露比例及胚存活之影響。 試驗一, 女豬經超級排卵後,分別在 hCG 注射後 52 ∼ 54 小時、54 ∼ 56 小時、56 ∼ 60 小時進行手術取胚, 比較不同時間取胚,所得 1- 細胞期胚數與其離心後原核顯露比例。 結果 1- 細胞期胚數與原核顯露比例分別在 52 ∼ 54 小時者為 6.9 及 31.4%、在 54 ∼ 56 小時者為 22.4 及 76.7%、在 56 ∼ 60 小時者 為 14.3 及 57.5%。試驗二,比較最大離心半徑下不同相對離心力( 0、14,900、20,600、 25,500、30,800、36,700 與 43,000 �T g ) 1- 細胞期豬胚之原核顯露比例與離心處理對 其存活率之影響。 結果不同相對離心力下其原核顯露比例分別為 0.0%、 51.4%、 75.0%、 79.3%、88.6%、87.0%、86.0%;其胚存活率則分別為 66.6%、51.4%、57.1%、41.3%、48.6% 、38.7%、與 37.2%。綜合上述結果顯示,1. hCG 注射後 54 ∼ 56 小時進行手術取胚,可 不宜超過至 25,500 �T g,可獲得較佳的原核顯露比例而不影響豬胚活率,相對離心力不宜 超過至 25,500 �T g,可獲得較佳的原核顯露比例而不影響豬胚的存活率。 |
英文摘要 | The purposes of this study were to find suitabe operation times to flush more pronuclear pig embryos and a suitable relative centrifugal field to expose fronuclei and not impair embryonic survial rate. Gilts were synchronized, superovulated, and artificially inseminated at 24 h and 30 h after hCG was injected. In the first experiment, three different operation times ( 52 to 54 h, 54 to 56 h, and 56 to 60 h after hCG injection) were compared to find the optimum time for recovering more 1-cell embryos and increasing pronucleus-exposured ratio (PER). Comparing the mean of 1-cell embryos recovered and PER, the operation of flushing held at 54 to 56 h (mean= 22.4, n=24; PER= 76.7%, 454/592) was better than at 52 to 54 h (mean=6.9, n=9; PER= 31.4%, 48/153) or 56 to 60 h (mean=14.3, n=38; PER=57.5%, 429/746) (9<0.05). In the second experiment, the effect of different relative centrifugal field (RCF) on PER was compared by centrifuging 1-cell embryos with 0, 14,900, 20,600, 25,500, 30,800, 36, 700, and 43, 000 �T g under maxium centrifuge radius. PER (0%, 51.4%, 75.1%, 79.3%, 88.6%, 87.0%, and 86.0%, respectively) increased as RCF was elevated and reached a plateau at 20,600 �T g. However, the embryonic developmental ability was significantly reduced by comparing 20,600 with 25,500 �T g (P<0.05). The results indicated that a donor operated on at 54 to 56 h, after hCG injection, would provide the most suitable stage of embryos for microinjection; and that RCF should bot be increated over 25,500 �T g to obtain better PER as embryonic survival rate was considered. |
