中文摘要 | 夜來香優良品系(77-A-05)之微體繁殖,採剛抽離地面之幼花序上<8mm的花蕾 為培植體,培養於MS修正培養基,含BA 4.5mg/l及NAA-0.1-0.2mg/l具最佳芽體再生。而 花梗節培植體,則於MS修正培養基,含BA 4.5mg/l,配合15-30天暗處理,可得最多量之 芽體。將6個優良品系之花蕾及花梗節培植體,培養於上述之培養基組合,進行初代培養 ,結果以花蕾為培植體者,品系間芽體形成率具78-100%的差異表現;花梗節培植體之芽 體形成率,品系間亦呈現從43-90%的差異。再生芽體數,以花蕾為培植體者,平均為2. 73個:花梗節培植體芽體平均為2.82個。將初代建立之芽體其繼代培養基將BA濃度提高至 6.75mg/1組合NAA 0.lmg/l,芽體增殖潛力亦大,6個品系平均繁殖效率達4倍以上。此法 採用之二種培植體,除了不破壞原有母株之生長與結球外,且材料取得與表面消毒容易, 繁殖倍率亦高,對選育之夜來香優良品系之大量繁殖,甚具應用價值。 |
英文摘要 | The florets (below 3 mm in length) and stalk node (5mm) of the young inflor escence were used as explant source for in vitro propagation of selected tuberose hybrid (77-A-05). A modified Murashige-Skoog medium containing 4.5mg/1 6-benzladenine purine (BA) and 0.1-0.2 mg/1 Naphthaleneacctic acid (NAA) was suitable for adventitious bud regencration. However, flower-stem-node explants cultured on a modified MS medium containing 4.5 mg/1 BA and placed in the dark for 15-30 days produced the greater bud number. Both young florets and flower-stem-nodes were proved satisfactory explant sources for six selected tuberose hybrids in primary culture. The result showed 78-100% of the floret explants and 43-90% of the flower-stem- node explants forming buds respectively. As for the number of bud induced from young florets and flower-stem-node 2.73 and 2.82 buds/explant, respectively. The established buds in primary explants were transferred to the same medium with 6.75 mg/1 BA and O.1mg/1 NAA for further shoot proliferation. An average 4 or more plantlets was produced in each subculture from six selected hybrids. Using young florets and flower-stem-nodes as explant sources for in vitro propagation of selected tuberose hybrids had several advantages: i). Growth and development maintenance of mother plant, ii). Easy to obtain explant sources, iii). Low contamination rate of explants. In vitro propagation technique using these two explants would be valuable for the rapid and large-scale multiplication from selected tuberose hybrids. |
