中文摘要 | 在豬放線桿菌胸膜肺炎的肺臟組織中,常可見到肺泡腔內出現大量排列成游渦狀 的多形單核細胞,為瞭解這些細胞之來源及其形成的過程, 本實驗以總菌量為 5 × 10 �� CFU 之胸膜肺炎放線桿菌,接種 8 隻無特定病原豬,於 6、12、24 及 48 小時之後,各犧 牲 2 隻,取出肺臟, 將有病變處以 10% 福馬林液固定,經石臘包埋,以切片機切成 6 μ m 之組織切片,再以伊紅及蘇木紫染色,用光學顯微鏡觀察豬肺泡腔內細胞形態之變化,同 時, 另以冷凍切片機將有病變處之組織切成 10 μ m 之切片,再以酸性磷酸�t細胞化學染 色法加以染色,用以證明肺泡內之多形細胞是否含有酸性磷酸�t,此外,也將有病變之組織 ,以 4% 戊二醛液固定,再以 1% 四氧化鋨進行後固定,經酒精脫水後,包埋於塑膠中,以 超薄切片機切成 75nm 薄片,用穿透式電子顯微鏡觀察肺泡腔內細胞之超顯微結構,以瞭解 其來源及特性。此外更以免疫細胞化學染色證明胸膜肺炎放線桿菌確實存在肺組織之病變處 。實驗結果顯示,豬感染胸膜肺炎放線桿菌之初期,出現在肺組織及肺泡腔內的炎症細胞是 以嗜中性球為主,但感染 24 小時以後,圓形細胞便成為本病最顯著的細胞形態,其中有些 細胞呈空泡化,核被推擠到細胞邊緣,當這種細胞大量聚集在肺泡腔內時,則堆積成游渦狀 的多形單核細胞,這些細胞經酵素細胞化學染色,結果顯示細胞質內具有酸性磷酸�t,因此 判定為巨噬細胞。以穿透式電子顯微鏡觀察也發現這些細胞的細胞質中,含有大量的溶素體 。由以上結果推論,在豬放線桿菌胸膜肺炎的病變內,肺泡腔之多形單核細胞,乃是由巨噬 細胞演變來的。 |
英文摘要 | The polymorphic mononulear cell (PMC) is an important finding of Actinobacillus pleuropneumoniae (App) infected lung in pigs. Although the characteristics of these cells had been studied, however, the process of formation is still unclear. IN this study, we had observed the serial pathological changes of alveolar cells, above all, the formation of PMC in pig's lung after App inoculation. Eight specific pathogens free pigs were inoculated intratracheally with App at a concentration of 5 X 10 �� CFU, then, each two were sacrificed at 6, 12, 24 and 48th hours later. The cells in the alveolar spaces were observed with light and transmission electron microscope, and analyzed cytochemically for acid phosphatase. The results revealed that a lot of neurtrophils were observed in the alveolar spaces at the early stage after inoculation. Twenty-four hours later, a lot of round cells appeared in lung tissue. Some of them became vacuolated, the nuclei were pushed into marginal area of cytoplasm, and some of them piled together to form a palisade or whirl arrangement. They were PMC. Enzyme cytochemical findings indicated that these cells were macrophages, because the acid phosphatase were strongly detected in their cytoplasm. Ultrastructural observation found that many lysosomes appeared in the cytoplasm of macrophages. Conclusively, it could be suggested that the PMC were derived from macrophages. |
