中文摘要 | 豬瘟是一種死亡率極高之豬隻惡性傳染病。本所擬利用遺傳工程方法開發一種利 用血清學方法即可將免疫豬與感染豬加以區別之本土性豬瘟次單位疫苗以進行豬瘟撲滅計畫 。 吾人從本省豬瘟病豬脾臟純化總 RNA,再利用 RT-PCR 方法合成 EO 與 E2 基因之 cDNA ,並將其選殖到細菌質體。本省分離株之 EO 與 E2 之核酸與蛋白質序列均與 Alfort 株 ( 法國豬瘟野外毒 ) 最為相似,其 EO 之同質性分別高達 92% 和 96%,而 E2 之同質性分別 為 89% 與 92%;然與本省以往所分離之豬瘟病毒 P97 株之同質性則相差較大。 |
英文摘要 | Hog cholera (HC)is a highly contagious and often fatal disease of pigs. To develop subunit vaccine with whcih pigs vaccinated can be serologically differenctiated from pigs that are infected with field strain, we cloned neutralizing antibody-inducing EO and E2 genes of HCV local strain. The total RNAs isolated from the spleen of a diseased pig were reversely transcribed and amplified by PCR with specific primers. The amplified EO and E2 cDNA were cloned into cloning vector. The cDNA sequences and deduced protein sequences of the EO and E2 were analyzed. They were closest to those of Alfort strain rather than those of P97 strain which had been isolated from Taiwan. The homology of the EO between these two strains was 92% and 96%, respectively; E2 89% and 92%, respectively. |
