中文摘要 | 本實驗針對馴養前後淡水長腳大蝦(Macrobrachium rosenbergii)肝胰臟內菌相進行研究。來自養殖場外觀健康的蝦子分成兩組,一組16隻,實驗前於30℃充分打氣的池水中馴養一週;另一組16隻蝦則不經馴養。結果證明馴養後蝦子肝胰臟中之總菌數比未經馴養者低兩個對數值,但兩者細菌組成相似,分離菌株中格蘭氏陽性菌分別佔38%及37%,格蘭氏陰性菌則佔62%及63%。60株格蘭氏陰性分離株進一步鑑定,顯示這些細菌共包括14個種,分屬於7個屬、4個科;其中Citrobacter freundii為優勢菌種,佔分離菌珠的41.7%。利用最低抑制濃度分析法發現,不同種的茵株對oxytetracy-cline-HCI及chloramphenicol兩種抗生素的敏感性不一。為了評估分離菌株是否具有致病潛力,本研究亦對分離株進行六種胞外產物活性測定,包括:糊精酵素(amylase)、脂肪酵素(lipase)、蛋白酵素(protease)、明膠酵素(gelatinase)、機丁質酵素(chintinase)及溶血素(hemolysin)。結果得知:(1)所有分離到的Aeromonas屬菌株都能產生五種胞外酵素,且具溶血性;(2)Pseudomonas aeruginosa可產生除了糊精酵素外的4種酵素,亦不具溶血性;(3)C. freundii則完全不具5種酵素活性與溶血性。我們相信,Aeromonas分離株和Pseudomonas aeruginosa的這些特性對淡水長腳大蝦可能具有致病潛力。 |
英文摘要 | This study is an investigation of bacterial flora found in the hepatopancreas (HP) of apparently healthy giant freshwater prawn, Macrobrachium rosenbergii. Prawns purchased from a local fish market were divided into two group, one group of sixteen prawns was acclimated in dechlorinated freshwater at 30℃ for one week prior to use in experiments, and the other group of sixteen unacclimated prawns was examined bacteriologically immediately following their acquisition. Total bacterial number in HP of the former was found to be two logs lower than in the latter, yet both groups of animals had similar bacterial compositions: of all isolated HP bacteria, 38% and 37% Gram-positive bacteria and 62% and 63% Gram-negative bacteria, 38% and 37% Gram-positive bacteria and 62% and 63% Gram-negative bacteria were found in acclimated and unacclimated pawn, respectively. Sixty strains of Gram-negative bacteria were further identified from four families, seven genera, and fourteen species; the predominant species was Citrobacter freundii, with an isolation rate reaching 41.7%. Using a minimum inhibitory concentration method, it was found that antibiotic resistance to oxytetracycline-HCI and chloramphenicol varied among several of the bacterial species. In order to assess the potential pathogenicity of isolates, hemolysis and the activities of five extracellular enzymes: amylase, lipase, protease, gelatinase, and chitinase were investigated. The results show that 1) the Aeromonas isolates produced all five extracellular enzymes and hemolysin; 2) Pseudomonas aeruginosa produced four of the five enzymes (less amylase) plus hemolysin; and 3) C. freundii failed to produce any of the extracellular enzymes or hemolysin. We believe that these characteristics might confer on the Aeromonas spp. And P. aeruginosa the potential to act as opportunistic pathogens in giant frewhs. |
