中文摘要 | 利用臺灣櫸之下胚軸為培植體在含1 ppm 2,4-D 之 WPM 固態培養基中誘導出白色 鬆軟癒合組織, 細切後置於含有 50 ml WPM 液態培養基之 125 ml 錐形三角瓶中振盪培養 ,每十天繼代培養一次,可建立穩定的細胞懸浮培養系統。以懸浮細胞做為原生質體分離材 料, 在 2% Cellulysin 及 0.5% Pectolyase Y-23 之酵素溶液處理下,可獲最大收量 3.4 × 10 �� pts/ml;原生質體經培養三天後仍有 80% 存活,於培養第七天可檢測到細胞壁再 生。另以葉片經泡水處理後,可於 Cellulysin 及 Macerozyme 溶液中分離出原生質體。 |
英文摘要 | Callus were induced from hypocotyls of Zelkova serrata which were cultured in solidified WPM medium containing 1 ppm 2,4-D, then they were chopped and inoculated into 125 ml flask containing 50 ml WPM liquid medium supplemented with 1 ppm 2, 4-D, and stable cell suspension culture were established after the flask were rotated with 125 rpm and subcultured every 10 days. Protoplasts were isolated from the suspension cultured cells by using enzyme slution containing 2% Cellulysin and 0.5% Pectolyase Y-23 with yield of 3.4 × 10 �� pts/ml. After 3-days incubation, about 80% of protoplasts were still viable and cell wall regenerated on the 7th day. Protoplasts were also isolated from leaves of Zelkova serrata after the leaves were pretreated with water and thenafter treated with enzyme solution containing Cellulysin and Macerozyme. |
